الوصف: Perceptual decision-making is a process of using available sensory information to select a response and guide behavior. Depending on the amount of relevant information and its reliability, the decision process may take more or less time to optimize the response. Indeed, if the information is ambiguous, delaying the choice execution is needed to gather more evidence. My PhD project aimed at understanding the mechanisms underlying such action regulation during decision-making. In this regard,, we were interested in the regulation of the activity of the striatum, a structure involved in action selection, exerted by the parvalbumin interneurons (PV). These PV interneurons receive many afferents from prefrontal cortical areas involved in decision processes. al cortical areas involved in decision processes. When activated, these interneurons exert strong inhibition on striatal projection neurons, modulating the overall striatal output. We hypothesised here that PV interneurons are recruited during perceptual decision-making. The higher the decision, the stronger the striatal inhibition by PV interneurons would have to be to delay the choice execution. We developed a novel freely-moving task of perceptual decision-making in mice enabling to precisely measure the duration of the decision process. This behavioural task is paired with fiber photometry recordings in order to monitor striatal PV interneurons activity along the task. The results described in this manuscript showed variation of the decision-making duration as a function of the mouse uncertainty context. As the mouse becomes expert in the task, the decision-making duration decreases suggesting an optimization of the decision process. Conversely, the higher the task difficulty, the longer the decision-making duration. Regarding the interneuronal activity, fiber photometry recordings showed a recruitment of PV interneurons during decision-making followed by a strong decrease of their activity specifically during their choice execution, suggesting a crucial role ...
