الوصف: Introduction: In general, human sperm is preserved in temperature range under -20°C. Currently, survival rate of the post-thaw sperm is around 50-60%. Although sperm cryopreservation medium including proteins and lipids derived from animals, for example egg yolk as a cryoprotectant, is commonly used, the components of proteins and lipids derived from animals are not clear. Therefore, synthetic and plant derived medium was used for the study. In the present study, post-thaw motility frozen with medium including long-chain alcohol and lipids was examined to develop cryopreservation medium for human sperm. Materials and Methods: Lauryl alcohol (LAl), a long-chain alcohol and Vitamin E (VE) were compared. Group (1): LAl 0-0.05% (v/v). Group (2): VE. 0-0.5% (v/v) and Group (3): medium with LAl and VE. LAl and VE were added to the medium compared in different concentrations and compared. The human sperm samples were made by adding the same volume of human sperm, adjusted to 100 million/ml to 0.5 ml sperm cryopreservation medium, and then thawed by liquid vapor method. The survival effect of human sperm was judged by the rate that showed mobility and was shown as sperm motility in percentage. Results: While the medium without LAl or VE in both Group (1) and (2) was 57.3%, in Group (1), LAl 0.02% added medium showed 66.5% sperm motility and in Group (2), VE 0.4% added medium showed 63.3% sperm motility. In Group (3), mixture of LAl 0.02% and VE 0.4% medium obtained 69.8% sperm motility. Conclusion: In the present study, LAl and VE as sperm cryoprotectants were evaluated, and 0.02% of LAl and 0.04% of VE in cryopreservation medium were effective on sperm motility. On the basis of the results, LAl and VE were considered safer, which would substitute alcohol and VE for generally-used components derived from animals. Furthermore, they give higher cryopretective effects for human sperm, and would work for actual human sperm cryopreservation and especially for cases with low number of sperm.
