الوصف: PURPOSE: The transcription factor nuclear factor-kappaB (NF-kappaB) is constitutively activated in most tumors, including gliomas. By virtue of its ability to regulate the expression of genes involved in apoptosis, differentiation, chemoresistance, migration, and survival, NF-kappaB constitutes the point of convergence of many oncogenic pathways. Recently, dehydroxymethylepoxyquinomicin (DHMEQ) has been shown to be a potent NF-kappaB inhibitor with antitumor and chemosensitizing properties. Nonetheless, despite the improvements in neurosurgery, radiation treatment, and the advent of temozolomide (TMZ), the outcome of patients with chemoresistant tumors is still poor. METHODS: The effects of NF-kappaB inhibition by DHMEQ (alone or combined with TMZ) were evaluated in pediatric glioblastoma (SF188) and pilocytic astrocytoma (R286) cell lines by means of proliferation (XTT assay), viability (Trypan blue exclusion), clonogenic capacity, and apoptosis (Nucview 488 caspase-3 kit) assays. Cells were treated with different concentrations (2.5, 5, 10 and 20 ug/ml) for 24, 48, and 72 hours. All experiments were performed in triplicate, and the results were compared using one-way analysis of variance followed by Bonferroni tests. RESULTS: DHMEQ treatment resulted in cell proliferation arrest in a dose- and time-dependent manner in both cell lines when compared with control. Cell viability and clonogenicity were also significantly diminished with the consequent increase of apoptotic cells. DHMEQ combined with TMZ showed synergy compared to TMZ treatment alone. CONCLUSION: Our results show the antitumor effect of DHMEQ and suggest that it is a good candidate for use as a new chemotherapeutic agent against gliomas.
