الوصف: | 93 Nandrolone is one of the synthetic anabolic steroids banned in sports. It has been shown that nandrolone metabolites including 19-norandrosterone (19-NA)、19-noretiocholanolone (19-NE) and 19-norepiandrosterone (19-NEA) are present in urine of various 19-norsteroids. In this study we investigated the conjugated moieties of the 19-NA、19-NE and 19-NEA in various 19-norsteroids. Urinary samples were obtained after healthy male volunteers orally administered one single dose (10 mg) of four 19-norsteroids. Enzymatic (��-glucuronidase from E. coli) and chemical (trimethyliodosilane; TMCS) hydrolyses were applied to deconjugate glucuro- and sulfo-conjugated metabolites before gas chromatography-mass spectrometry (GC-MS) analysis. Results from this study showed that (1) 19-NA and 19-NE were present as both glucuronide and sulfate conjugates; (2) 19-NEA was excreted exclusively as sulfate conjugates; (3) the concentration profiles of 19-NA, 19-NE and 19-NEA metabolites in urine reached peaks between 2 to 12 h post-administration and declined thereafter until approximately 72 to 96 h; (4) a greater amount of glucuro-conjugated metabolites were present at the early stage of excretion; (5) at the later stage of excretion, however, various amounts of sulfo-conjugated metabolites were present in all the steroids analyzed; (6) amount of 19-NA was generally higher than 19-NE in most compounds analyzed; (7) although rare, 19-NE may show higher amount than 19-NA during the course of excretion; and (8) prominent inter-individual variability was observed in the excretion of the metabolites tested in this study. |
