    الوصف: Vaccinium oldhamii Miq. is an edible berry; however, little is known about its seed dormancy-break and germination. Therefore, the aim of this study was to elucidate the seed-dormancy break and germination characteristics of V. oldhamii. The results showed that the length and width of the seeds of V. oldhamii were 2.4588 ± 0.0273 mm and 1.7028 ± 0.0248 mm, respectively, the filled percentage was 49.0 ± 3.0%, and the 1000-seed weight was 0.9453 g. Additionally, water imbibition test showed that the seed weight of V. oldhamii increased by more than 70% after 24 h, indicating the absence of physical dormancy. Embryo length measured at dispersal and just before germination did not differ significantly. There was no significant difference in E:S (Embryo:Seed) ratio in length at the time of seed dispersal (0.6780 ± 0.0258) and before germination (0.7370 ± 0.0469), indicating the absence of morphological dominance. Temperature treatments showed that the germination of the seed was 0, 6.0, 6.0, 20.0, and 0% under 15, 20, 25, 25/15, and 15/6 °C, respectively. However, treatment with 250 ppm of GA3 increased the germination of the seeds to 22.0, 36.0, 32.0, 40.0, and 1.0%, respectively. Additionally, treatment with 500 ppm of GA3 increased the germination to 34.6, 40.0, 40.0, 46.0 %, respectively. These results indicated that the seeds of V. oldhamii exhibited Nondeep physiological dormancy (Nondeep PD). Overall, the findings of this study showed that treatment with 500 ppm of GA3 at 25/15 °C can effectively break the dormancy of V. oldhamii seeds, which could facilitate further research on the species.

    الوصف: Abstract Background Vaccinium oldhamii (V. oldhamii) has been reported to exert a variety of the pharmacological properties such as anti-oxidant activity, anti-cancer activity, and inhibitory activity of α-amylase and acetylcholinesterase. However, the anti-inflammatory activity of V. oldhamii has not been studied. In this study, we aimed to investigate anti-inflammatory activity of the stem extracts from V. oldhamii, and to elucidate the potential mechanisms in LPS-stimulated RAW264.7 cells. Methods Cell viability was evaluated by MTT assay. The determination of NO and PGE2 production was performed using Griess reagent and Prostaglandin E2 ELISA Kit, respectively. The change of mRNA or protein level was evaluated by RT-PCR and Western blot. Results Among VOS, VOL and VOF, the inhibitory effect of NO and PGE2 production induced by LPS was highest in VOS treatment. Thus, VOS was selected for the further study. VOS dose-dependently blocked LPS-induced NO and PGE2 production by inhibiting iNOS and COX-2 expression, respectively. VOS inhibited the expression of pro-inflammatory cytokines such as IL-1β, IL-6 and TNF-α. In addition, VOS suppressed TRAP activity and attenuated the expression of the osteoclast-specific genes such as NFATc1, c-FOS, TRAP, MMP-9, cathepsin K, CA2, OSCAR and ATPv06d2. VOS inhibited LPS-induced NF-κB signaling activation through blocking IκB-α degradation and p65 nuclear accumulation. VOS inhibited MAPK signaling activation by attenuating the phosphorylation of ERK1/2, p38 and JNK. Furthermore, VOS inhibited ATF2 phosphorylation and blocked ATF2 nuclear accumulation. Conclusions These results indicate that VOS may exert anti-inflammatory activity by inhibiting NF-κB and MAPK/ATF2 signaling. From these findings, VOS has potential to be a candidate for the development of chemopreventive or therapeutic agents for the inflammatory diseases.

    الوصف: The root diameters as well as colonization and diversity of the root-associating fungi of Vaccinium oldhamii Miq. were investigated in order to obtain information on their mycorrhizal properties. The distal regions of roots had typical hair roots with diameters of less than 100 μm. Ericoid mycorrhizal fungi (ErMF) and dark septate endophytes (DSE) were frequently observed in the roots. Ascomycetes, particularly helotialean fungi, appeared to be dominant among the endophytic fungi of V. oldhamii roots. Furthermore, Rhizoscyphus ericae (Read) Zhuang & Korf and Oidiodendron maius Barron known as ErMF were detected more frequently than other fungal species.

    الوصف: 국내 자생 정금나무의 기능성 품종을 육성하기 위하여 4집단 24개체에 대한 열매의 형태적 특성과 안토시아닌 함량을 분석하였다. 분산분석결과 조사된 모든 형질 특성들에서 집단간 및 집단내 개체간에 유의적인 차이가인정되었다. 정금나무 열매의 평균특성을 살펴보면 열매종경 8.2 mm, 열매횡경 7.8 mm, 열매무게 0.4 g, 열매당도11.7 brix., 과지당 열매수 7.7개, 안토시아닌 함량 474.4 mg/100 g으로 각각 나타났다. 특히, 정금나무의 열매특성은미국산 블루베리에 비해 크기는 작으나 안토시아닌 함량이 1.9배 높은 것으로 나타나 기능성 품종육성을 위한 소재로서의 이용 가치가 높은 수종으로 판단된다. 기능성 품종육성을 위하여 3가지(열매무게, 과지당 열매수, 안토시아닌함량) 주요 선발기준에 의해 최종 선발된 3개체의 선발효과를 추정한 결과 전체 평균특성과 비교하여 열매무게129.6%(0.36 g→0.47 g), 과지당 열매수 109.5%(7.7개→8.4개), 안토시아닌 함량 140.5%(474.4 mg/100 g→666.4 mg/ 100 g)의 선발효과를 추정할 수 있었다.
