    الوصف: Fluorine holds a prominent position within the realm of drug discovery and development, substantiated by its presence in approximately 25% of drugs approved by the US Food and Drug Administration (FDA). Consequently, the advancement of new fluorination reactions stands as a pivotal area in medicinal chemistry. In particular, the monofluoro-, difluoromethyl-, and trifluoromethyl- are three groups that appear most frequently in drug structure. Quinoline, owing to its privileged structural status, plays a crucial role in drug design and synthesis. Various approaches have been documented for the direct difluoromethylation of the C-2 and C-4 positions of the quinoline ring. However, achieving direct C-3 difluoromethylation has remained an elusive objective. In this study, we introduce a novel method for effecting the direct difluoromethylation at the C-3 position of the quinoline ring. Comprehensive characterizations, including 1H-NMR, 13C-NMR, and 19F-NMR for all compounds are performed. We believe that this novel method will open a new way to access the hitherto untapped C-3-difluoromethylation active compounds.

    الوصف: Abstract The urban mass rapid transit (UMRT) Line HN2A is the first light rail transit line in Vietnam. It is also the first time the operational safety assessment for the whole life cycle of a railway project is applied and assessed by an applicable scientific tool. While various industry standard methods have been deployed in many countries, their application is not appropriate for assessing the outdated railway infrastructure in Vietnam. This article proposes a method for generating safety risk models for train collisions using the fault tree analysis (FTA) technique. The FTA method comprehensively evaluates the fundamental error and failure probability that could potentially lead to accidents in general and train collisions in particular on Line HN2A. The study describes the procedure for establishing FTA and determining assumptions based on technical specifications and similar railway systems. Compared with the statistical failure results using data from operational tests and commissioning (2018–2021) of the metro line, the results here indicate that this is a reasonable theoretical model applicable to UMRT in Vietnam. The theoretical model will be processed to generate the first-ever scientific risk assessment system based on empirical evidence. In addition, real-time accident and operation data will continue to be collected and compared to the theoretical model to improve its accuracy. The findings of this study could serve as a starting point for risk management on current and future freight, passenger, and metro lines in Vietnam.

    الوصف: Aims: The study was conducted on black soybean seeds (Glycine max. L) to optima the conditions of some factors affecting the extracted yield of polyphenol compounds. Study Design: The factors include the types of solvent used (methanol, ethanol and acetone); solvent concentrations (40, 50, 60, 70, 80 and 90 % v/v); ratio of sample and solvent (1:4,1:6, 1:8, 1:10) and cycle of extractions (2, 3, 4); extract times (2, 3, 4h) and temperature (30, 40, 50, 60 oC). The efficiency of polyphenol extract was shown by the content of total phenolic (TPC) and flavonoid (TFC). Place and Duration of Study: Department of Biotechnology and Food technology, Thai Nguyen University of Agriculture and Forestry, Viet Nam, from March 2021 to March 2022. Methodology: The black soybean seeds were pulverized by a mill and defatted using the Soxhlet system with hexane for 10 hrs as described by Weidner et al. (2012). TPC was determined by the Folin-Ciocalteu method of Susu Giang et al (2013). TFC was determined by colorimetric method as described by Ozsoy et al (2008). Results: The results showed that the extraction efficiency was high when using 70% acetone solvent; the appropriate ratio of soybean and solvent was 1:10 with 3 cycles of extraction. The extraction efficiency can be improved when extracting samples at temperature at 60oC for 4 hours, TPC and TFC content reached about 5.02±0.04 mgGAE/g and 2.74±0.03 mgQE/g. Conclusion: 70% acetone is the most suitable solvent to extract polyphenols from black soybean. The ratio of sample in solvent is 1:10 (w/v), three times of extraction in 4 hours for each extraction cycle at 60°C for the highest extraction efficiency of TPC and TFC, reaching 7.02 ±0.04 mgGAE/g and 2.74±0.03 mgQE/g, respectively.

    الوصف: Herein, we report the design, synthesis and evaluation of novel ( E )‐3‐(3‐oxo‐4‐substituted‐3,4‐dihydro‐2H‐benzo[b][1,4]oxazin‐6‐yl)‐ N ‐hydroxypropenamides ( 4 a – i , 7 a – g ) targeting histone deacetylases. Three human cancer cell lines were used to test the cytotoxicity of the synthesized compounds (SW620, colon; PC‐3, prostate; NCI−H23, lung cancer); inhibitory activity towards HDAC; anticancer activity; as well as their impact on the cell cycle and apoptosis. As a result, compounds 4 a – i bearing the alkyl substituents seemed to be less potent than the benzyl‐containing compounds 7 a – g in all biological assays. Compounds 7 e – f were found to be the most active HDAC inhibitors with IC 50 of 1.498±0.020 μM and 1.794±0.159 μM, respectively. In terms of cytotoxicity and anticancer assay, 7 e and 7 f also showed good activity with IC 50 values in the micromolar range. In addition, the cell cycle and apoptosis of SW620 were affected by compound 7 f in almost a similar manner to that of reference compound SAHA. Docking assays were carried out for analysis the binding mode and selectivity of this compound toward 8 HDAC isoforms. Overall, our data confirmed that the inhibition of HDAC plays a pivotal role in their anticancer activity.

    الوصف: Eliminating latently infected cells is a highly challenging, indispensable step toward the cure for HIV/AIDS. Hypothesis put forward herein is that the unique HIV protease cut site (Phe-Pro) can be reconstructed using a potent inhibitor of tubulin polymerization, monomethyl auristatin F (MMAF), which features Phe at its C-terminus. This presents opportunities to design prodrugs that are specifically activated by the HIV protease. To this end, a series of MMAF derivatives is synthesized and evaluated in cell culture using latently HIV-infected cells. The cytotoxicity of compounds is indeed enhanced upon latency reversal by up to 11-fold. As a result, in a mixed cell population, nanomolar concentrations of the lead compounds depletes predominantly the HIV-infected cells and in doing so markedly enriches the pool of the uninfected cells. Affinity of the lead compounds to the viral protease is validated computationally and experimentally but despite expectations, the mechanism of action of the synthesized toxins is shown to be independent from the enzymatic activity of the HIV protease.

    الوصف: Background COVID-19 vaccines have been speedily developed and deployed. The more vaccine doses are delivered to users, the more common adverse events following immunization (AEFI) are reported. This study aimed to identify factors affecting AEFI in Vietnamese people receiving the COVID-19 vaccine AZD1222 developed by AstraZeneca and Oxford University. Methods In July 2021, an online cross-sectional survey was conducted among Vietnamese who have been vaccinated with COVID-19 vaccines. The questionnaire collected demographic characteristics, medical history, types of injected vaccines, common AEFI, and post-vaccination activities from respondents. The effects of host-related factors on AEFI including 24 specific symptoms were also explored. Results After screening, 1028 participants who were Vietnamese, over 18 years old and received at least one dose of AZD1222, were included in the study. Only 40/1028 (3.9%) participants reported not having any AEFI, whereas 25/1028 (2.4%) reported to have severe symptoms. The most common AEFI were moderate fever (69.4%), muscle aches (68.6%), followed by fatigue/ sleepiness (62.5%), body aches (59.4%), headache (58.5%), pain at injection site (58.3%) and chills (45.7%). Data analysis showed that females complained about AEFI particularly gastrointestinal symptoms more frequently than males. Age of participants and number of doses were also important factors affecting AEFI as the increase of age or number of vaccine doses was associated with the decrease of self-reported AEFI frequency. Conclusions This study provides a detailed assessment of risk factors associated with AEFI in Vietnamese people vaccinated with AZD1222. It seems that gender, age and vaccine doses are important factors affecting AEFI.
