    الوصف: Umbilical cord blood (UCB) represents a source for hematopoietic stem cells (CB-HSC), and transplantation of cord blood has been part of clinical practice since more than 10 years (Lee et al., Blood 103, 2004). The UCB is usually collected by public or private cord blood banks, which store it for an unknown recipient for an undetermined time. The cryopreservation process, used for stem cell collection, is particularly critical for umbilical cord blood (Berz et al., Am J Hematol 82, 2007). The most used cryoprotectant agent (CPA) to avoid or reduce cell damage caused by ice crystals, is 10% dimethyl sulfoxide (Me2SO). Me2SO, while protecting the cells from membrane rupture, however has a toxic effect on them, depending on the temperature and the exposure time for both pre-freeze and post-thaw periods (Berz et al., Am J Hematol 82, 2007). Generation of oxygen-free radicals (ROS) is a major cause of cell damage during low-temperature storage (Katkova et al., Cryobiology 37 and Kearney, Burns 24, 1998). Cells counteract ROS accumulation through antioxidant enzymes, such as superoxide dismutase (SOD) and glutathione peroxidase (GPX). The addition of antioxidants to the conventional freezing medium improves the post-thaw cell recovery (Limaye et al., Stem Cell Res. 10, 2001). The purpose of this study is the improvement of cord blood HSC’s (CB-HSC) cryopreservation method by addition of a new isoform of recombinant manganese superoxide dismutase (rMnSOD), obtained from the native liposarcoma derived form (LSA-MnSOD) (Mancini et al., Int. J. Cancer 119, 2006). rMnSOD, while having the same enzymatic activity common to all SODs, differs for its higher molecular weight (30 kDa versus 24 kDa) due to the presence of an additional sequence, called leader peptide, not cleaved during its maturation. As demonstrated by Mancini et al. (2006), recombinant LSA-type MnSOD (rMnSOD) has a distinctive capacity to penetrate cells and converts ROS to H2O2.     الوصف: Manganese superoxide dismutase (MnSOD) is a mitochondrial enzyme that defends against oxidative damage due to reactive oxygen species (ROS). A new isoform of MnSOD with cytotoxic activity was recently discovered in liposarcoma cells. Here, we tested the effectiveness of a recombinant form of this isoform (rMnSOD) on leukemic T cells, Jurkat cells, and lymphocytes. Our results confirm that leukemic T cells can internalize rMnSOD and that rMnSOD causes apoptosis of 99% of leukemic cells without showing toxic effects on healthy cells. Using light and electron microscopy, we determined that an rMnSOD concentration of 0.067 μM most effective on apoptosis induction. Western blot analysis showed that treatment with 0.067 μM rMnSOD resulted in high expression of the pro‐apoptotic protein Bax and low expression of the anti‐apoptotic protein Bcl‐2 in leukemia cells. Concerning signal transduction pathway no influence was observed after treatment except for Jurkat cells showing a slightly decreased expression of ERK phosphorylation.     الوصف: The effectiveness of the filters available for the Novasina Hygrometer for measurement of water activity (aw) in intermediate moisture foods, most of them suspected of being potential sources of sensor contamination, was investigated. Foods studied were apple preserve, chocolate topping, honey, milk jam, salted anchovy, intermediate moisture beef, raw ham, dry salami, smoked pork sausage, provolone and roquefort cheese, tomato ketchup, semimoist pet food, concentrated tomato paste and Worcestershire sauce. For most foods, the use of an adequate filter permitted an acceptable precision in aw measurement; however, neither of the filters studied allowed measurement of aw in the pet food containing propylene glycol. ; Fil:Pollio, M.L.     الوصف: The aim of this paper is to establish similarities or differences between water sorption characteristics of the most important Argentine wheat varieties and to propose a statistical methodology for their comparison. Ten varieties of wheat were studied by an isopiestic method at 25°C. A balanced two-way analysis of variance with interactions was used. The usual F-tests, assuming normality of the errors, and free-distribution tests were carried out. For the latter test specific software was developed. Transformations were eventually made in order to verify the underlying assumptions of the model.     الوصف: A theoretical approach, based on the thermodynamic properties of strong electrolyte aqueous solutions, was used to calculate the aw at 5°C and 10°C of selected saturated salt solutions frequently used as standards in the range of microbial growth. The results agreed very well with experimental measurements at both temperatures for most salts studied, which included NaBr, NaCl, SrC12, (NH4)2SO4, KCl, BaCl2, and K2SO4.     الوصف: A re-evaluation of the water activity (aw) of saturated solution of KNO3 was performed in view of the discrepancy which exists on its exact aw value. Four different brands of KNO3 were used and ten determinations of aw were made for each one. The resulting mean value and its confidence interval (95% level) was 0.927 ± 0.001. This result, which agrees with a previous theoretical prediction of aw in saturated KNO3, indicates that the value 0.936 used by many authors should no longer be used. © 1988 Academic Press Limited. ; Fil:Pollio, M.L.     الوصف: Water adsorption isotherms at 25°C of five soybean varieties grown in Argentina were determined; oil contents ranged from 16.7 to 22.7% (dry basis). Equilibrium moisture curves of the different varieties were coincident when the moisture contents were calculated on an oil‐free basis. Water adsorption by soybean oil was negligible compared with the water adsorbed by the non‐oil fraction at the same relative humidities.
