    الوصف: Bioprospecting sub-explored environments such as Antarctic locations leads to finding out diverse activities, reducing harmful chemical usage that affects both human health and the environment. In this study, ~ 7000 cold-adapted bacterial strains were isolated from samples around Melchior Antarctic Base at 5 °C and more than 13,000 at 15 °C. Out of them, 900 different colony morphotypes were evaluated for antimicrobial production, and 13 isolates demonstrated antibacterial and antifungal activities. One isolate, closely related to Burkholderia gladioli according to 16S rDNA (99.8%), gyrB (99.6%) and Cpn60 (99.4%) gene sequence analysis, showed a consistent, broad antimicrobial spectrum against both pathogenic and phytopathogenic bacteria. Its potent antifungal activity inhibits the growth of various plant pathogenic fungi, whereas it was mainly studied against Penicillium digitatum and Macrophomina phaseolina, the causal agents of blue mould in postharvest fruits and charcoal rot in soybean crops, respectively.     الوصف: Background In pregnant women Streptococcus agalactiae (GBS) can be transmitted to newborn causing severe infections. It is classified into 10 serotypes (Ia, Ib, II-IX). The severity of neonatal disease is determined by the capsular serotype and virulence factors such as the polysaccharide capsule, encoded by the cps gene, protein C, which includes the Cα surface proteins (bca gene), Rib (rib gene) and Cβ (bac gene); the proteins Lmb (lmb gene), FbsB (fbsB gene), FbsA (fbsA gene), the cyl operon encoding a β-hemolysin (hylB gene), the CAMP factor (cfb gene) and the C5a peptidase (scpB gene). The aim of this work was to determine the degree of GBS colonization in pregnant women, the serotypes distribution and to investigate virulence-associated genes. Methods We worked with 3480 samples of vagino-rectal swabs of women with 35–37 weeks of gestation. The identification of the strains was carried out using conventional biochemical tests and group confirmatory serology using a commercial latex particle agglutination kit. Two hundred GBS strains were selected. Their serotype was determined by agglutination tests. The monoplex PCR technique was used to investigate nine virulence-associated genes (cps, bca, rib, bac, lmb, fbsB, fbsA, hylB and scpB). Results The maternal colonization was 9.09%. The serotypes found were: Ia (33.50%), III (19.00%), Ib (15.50%), II (14.00%), V (7.00%) and IX (5.50%). 5.50% of strains were found to be non-serotypeable (NT). The nine virulence genes investigated were detected simultaneously in 36.50% of the strains. The genes that were most frequently detected were scpB (100.00%), fbsA (100.00%), fbsB (100.00%), cylB (95.00%), lmb (94.00%) and bca (87.50%).     الوصف: The aim of this work was to determine the susceptibility, molecular profile, and clonal relationship in Streptococcus agalactiae (group B Streptococcus [GBS]) isolated from vaginal-rectal swab samples. We worked with 200 isolates collected from pregnant women between 35 and 37 weeks of gestation. The macrolide-lincosamide-streptogramin B (MLSB) resistance phenotypes were determined using the double-disc assay. Susceptibility to erythromycin (ERI) and clindamycin (CLI) was performed with the E-test. Resistance genes ermB and ermTR were detected by polymerase chain reaction. Clonal studies were performed using the random amplification of polymorphic DNA. Twelve (6%) of the isolates were resistant to ERI and 10 (5%) of them to CLI. Fifty percent of the resistant strains corresponded to serotype III, 25% to serotype V, and the remaining 25% to serotype Ia, II, and nontypeable strains. The cMLSB phenotype was detected in eight strains (66.67%) and the iMLSB phenotype in four (33.33%). The minimum inhibitory concentration values were between 1.5 and 16 μg/mL for ERI, and between 1 and 32 μg/mL for CLI. Out of the 25 strains susceptible to ERI and CLI, the presence of the ermB gene was detected in eight of them and the ermTR gene in one strain. The ermB gene was detected in the 12 strains that initially had some macrolide resistance phenotype. The ermTR gene was detected in three out of the four strains with the iMLSB phenotype. The resistance to macrolides in the province of Misiones is due to multiclonal spread. The phenotypic and genotypic characterization of macrolide resistance in GBS strains are crucial to contribute to the correct intrapartum prophylactic antibiotic therapy of allergic pregnant women and the epidemiological surveillance of these strains.     الوصف: In the context of a bioprospection programme for tyrosinase/L-DOPA- and melanin-producing fungal strains for biotechnological purposes, a hyperproducer isolate was obtained from Las Yungas rainforest, a relevant biodiverse ecoregion in North-Western Argentina. The selected strain was preliminarily identified as Paraboeremia sp. This is, to the best of our knowledge, the first native reported species of this genus in South America. Single-gene and multi-locus analyses of the internal transcribed spacer nuclear ribosomal RNA gene region (ITS), partial large subunit 28S nrDNA region (LSU), RNA polymerase II region (RPB2) and partial β-tubulin gene (TUB2) alignments were carried out to define the phylogenetic identity of this strain. As part of a polyphasic identification approach, these results were combined with morphological studies of active cultures growing on malt extract, oatmeal and potato dextrose agar plates. Incubation was performed under diverse conditions to stimulate sporulation for the subsequent micromorphological analysis. Microphotographs of pycnidia and conidia were taken with a scanning electron microscope. Maximum likelihood and Bayesian Inference analyses supported the location of the strain within the genus Paraboeremia, whilst morphological features allowed distinguishing it from previously described species within this genus. Based on the results herein reported, the new South-American species Paraboeremia yungensis is described and proposed.

    الوصف: Scleroglucan is a β-glucan exopolysaccharide (EPS) efficiently produced by Sclerotium rolfsii ATCC 201126, with attractive properties for a wide range of industries. Its production was herein comparatively assessed with nine alternative C- and twelve N-sources. When comparing conventional sucrose-based Production Medium PM20 (8.41 g C/L + NaNO3 as N-source) at shake-flask-scale vs. alternative C-source versions, soluble starch and sugarcane molasses led to efficient EPS production. At bioreactor scale, starch-based medium led to highest EPS production (7.95 g/L), recovery (~ 52%) and productivities (0.11 g EPS/L h; 0.018 g EPS/gbiomass h). Molasses, though leading to lower EPS production (5.11 g/L), could be envisaged as a promising agroindustrial sub-product for adding value and innovation. Oxalate side-product varied with C- and N-sources, with no clear detrimental relationship with EPS production. Agroindustrial sub-products showed then to be suitable as alternative substrates for efficient, low-cost, and scalable EPS production, thus opening new perspectives for medium reformulation and sustainable EPS production.     الوصف: In recent years, there has been an increasing interest in the remediation of contaminated environments, and a suitable solution is in situ bioremediation. To achieve this, large-scale bacterial biomass production should be sustainable, using economic culture media. The main aim of this study was to optimize the physicochemical conditions for the biomass production of an actinobacterium with well-known bioremediation ability using inexpensive substrates and to scale-up its production in a bioreactor. For this, the growth of four strains of actinobacteria were evaluated in minimal medium with glucose and glycerol as carbon and energy sources. In addition, l-asparagine and ammonium sulfate were assayed as alternative nitrogen sources. The strain Streptomyces sp. A5 showed the highest biomass production in shake-flasks culture using glycerol and ammonium sulfate as carbon and nitrogen sources, respectively. Factorial designs with five factors (glycerol concentration, inoculum size, pH, temperature, and agitation) were employed to optimize the biomass production of Streptomyces sp. A5. The maximum biomass production was obtained using 5 g L(−1) of glycerol, 0.25 µL of inoculum, pH 7, 30 °C and 200 rpm. Finally, the production was successfully scaled to a 2 L stirred tank bioreactor.     الوصف: Heavy metals at elevated concentrations are a major threat to agricultural and human health. Typically, human activities tend to release these metals to the environment in aqueous solutions, generating high levels of pollution due to the mobility of the heavy metals. The aim of the present work was to assess heavy metal tolerance in yeasts isolated from Rio Agrio - Lake Caviahue volcanic acidic aquatic environment and to evaluate the capacity of selected strains to capture metals in acidic culture media conditions. The ability of three yeast species, Cryptococcus agrionensis, Cryptococcus sp. 2, and Coniochaeta fodinicola, to tolerate and capture metals in live cultures has been evaluated. These three yeast species showed high tolerance to low pH and elevated concentrations of metals, thus implying their autochthonous status. Minimal inhibitory concentration (MIC) for growth obtained for these isolates showed elevated tolerance to the six heavy metals evaluated and were significantly higher than those registered for other microorganisms. C. agrionensis was able to capture 15.80 mg (g biomass)-1 of Cu2+ (MIC: 0.22 g L-1 ), Cryptococcus sp. 2 was able to capture 36.25 and 65.28 mg (g biomass)-1 of Ni2+ and Zn2+ , respectively (MIC: 0.56 and 1.68, respectively), and C. fodinicola was able to capture 67.11 mg (g biomass)-1 of Zn2+ (MIC: 3.75). This work reported the ability of yeasts to capture metals in acidic conditions for the first time. We hope that it represents the step-stone for future researches in the ability and metabolism of yeasts form acidic aquatic environment related to metal tolerance and capture.

    الوصف: Intensive efforts are being made to find new compounds with antimicrobial activity. The search for these novel bio-products in sparsely explored environments may be the key to providing solutions for many emerging problems. Antarctic environments are valuable locations for bioprospecting. In this study, 63 cold-adapted bacterial strains of 6100 different colony morphotypes were isolated from Antarctic seawater samples around South Shetland and Deception islands. Strains were selected based on cold-active antimicrobial production and were grouped into 11 operational taxonomic units by internal spacer region-PCR and amplified ribosomal DNA restriction analysis. Isolates arbitrarily named 2D, 5D, and 6D were closely related to Halomonas titanicae, with 99.8, 98.9, and 96.7% identity according to 16S rDNA sequencing, and 99.7, 99.3, and 98.3% according to gyrB region sequence analysis, respectively. The isolate 18SH was closely related to Candida sake (99.2%) based on sequence analysis of the ITS1?5.8S rDNA?ITS2 and D1/D2 domain of 26S rDNA.     الوصف: The aims of the present work were to isolate and identify the principal yeasts present in spoiled Argentine strawberry juice, identify polyphenols present in four blueberry cultivars and use these blueberry extracts in the control of yeasts using strawberry juice as food system model. Hanseniaspora osmophila and Starmerella bacillaris were identified for the first time in Argentine strawberry juice. The blueberry extracts assayed showed antifungal activity against H. osmophila and S. bacillaris through individual phenolic compounds such as quercetin, kaempferol and chlorogenic, ρ-coumaric and ellagic acid. The cytotoxicity assay demonstrated that the blueberries were not toxic to humans and that they did not modify the sensorial qualities of strawberry juice. No viable S. bacillaris and H. osmophila cells were detected after 7 days in strawberry juice supplemented with 150 µg/ml Blue Crisp or Millennium extract, inoculated with the isolated spoilage yeasts and conserved at 4 °C. This is the first evidence of S. bacillaris and H. osmophila in spoiled Argentine strawberry juice and blueberry extracts could be a good natural and non-toxic alternative to prevent growth of these yeasts. Blueberry extracts could be feasible alternatives to improve the microbiological quality without impact on the organoleptic properties of polyphenol-enriched strawberry juice.
