    الوصف: Certain mouse models of Cystic fibrosis (CF) exhibit a severe intestinal phenotype, resulting in death soon after birth or at the time o f weaning. Marked variations in intestinal disease severity among different CF mouse models have suggested a prominent role for secondary genetic modifiers ofthe disease. Calcium-activated, chloride channels (CLCA) have been postulated as genetic modifiers of CF. It has been previously shown that mCLCA3, a secreted protein involved in mucus production, shows markedly reduced expression in goblet cells o f the intestines o f a CF mouse model, and when expression is corrected through transgenic manipulation, can significantly ameliorate the disease. Current studies were undertaken using quantitative RT-PCR to show that expression of mClca6 is significantly reduced (p<0.005) in the ileum of a juvenile CF mouse model. Further quantitative RT-PCR studies were performed on the mildly-affected adult CF mice, showing significant differential expression of mClca6 in the jejunum (p<0.005), but not in the ileum. Moreover, a CF mouse model which exhibits relatively mild lung and intestinal disease compared to other mouse models shows no significant differential expression of mClca6 in the ileum. Although the function of mCLCA6 remains unclear, these results provide support for it as a potential modifier of intestinal disease severity

    الملخص (بالإنجليزية): Objective: Clinical scar assessment lacks standardized methodology and consensus on the most appropriate evaluation instrument. This study empirically evaluated whether area scars could be validly assessed by naive observers with the objective to develop and validate a novel multidimensional visual analogue scale (VAS) for the assessment of area scars. Methods: Standardized digital photographs of radial forearm free flap (RFFF) donor sites were obtained. Naive observers evaluated the images in three sequential psychophysical experiments, which led to the development of the new scar scale. These experiments involved initial evaluation of four dimensions (pigmentation, vascularity, observer comfort, acceptability) using a paired comparison (PC) paradigm and correlation with ratings of overall severity using a VAS, and initial VAS test phase followed by formal debriefing, and, subsequently, evaluation of a VAS for the four dimensions in addition to contour. Validation involved determination of intra- and interrater reliability and correlational analysis. Results: Across all three experiments, 56 observers evaluated 101 images, generating 12 720 observations for analysis. PC data demonstrated that observers could assess scars with high reliability and internal consistency for all dimensions (>95%). Overall (VAS) severity correlated highly with all dimensions, including contour. The new VAS yielded high levels of correlation (r = .72-.98, p < .01). Conclusion: Comprehensive VAS analysis demonstrates high reliability in mirroring PC results for multiple dimensions of area scars. These data support our novel multidimensional VAS method as a valid, reliable, simple, and time-efficient instrument for clinical and research use.     Abstract (French): But: L'évaluation clinique des cicatrices manque d'uniformité et ne fait pas consensus en ce qui concerne les instruments d'évaluation les plus pertinents. La présente étude visait à vérifier, de façon empirique, si l'évaluation de cicatrices par des observateurs non expérimentés pouvait donner lieu à une appréciation valable des cicatrices en vue de l'élaboration et de la validation d'une nouvelle échelle visuelle analogue (EVA) pluridimensionnelle d'évaluation des cicatrices. Méthode: Nous avons obtenu des photographies numériques, normalisées de zones donneuses de lambeaux libres prélevés sur la face externe de l'avant-bras. Les observateurs non expérimentés ont évalué les images dans trois expériences psychophysiques séquentielles, qui ont mené à l'élaboration de la nouvelle échelle d'évaluation des cicatrices. Les expériences comprenaient une évaluation initiale de quatre aspects, soit la pigmentation, la vascularisation, le degré d'aise de l'observateur et l'acceptabilité, fondée sur un paradigme de comparaison appariée (CA) et une corrélation des cotes de gravité globale à partir d'une EVA, une phase d'essai initial de l'EVA, suivie d'une séance structurée de déconditionnement et, par la suite, une évaluation d'une EVA sur les quatre aspects précédents ainsi que sur les bords. La validation comprenait la détermination de la fidélité intra- et interobservateurs ainsi que l'analyse des corrélations. Le degré de gravité globale sur l'EVA était en étroite corrélation avec tous les aspects examinés, y compris avec les bords. La nouvelle EVA a été associée à des degrés élevés de corrélation (r = 0.72 - 0.98; P < 0.01).
