الوصف: BACKGROUND: Codon usage and codon-pair context are important gene primary structure features that influence mRNA decoding fidelity. In order to identify general rules that shape codon-pair context and minimize mRNA decoding error, we have carried out a large scale comparative codon-pair context analysis of 119 fully sequenced genomes. METHODOLOGIES/PRINCIPAL FINDINGS: We have developed mathematical and software tools for large scale comparative codon-pair context analysis. These methodologies unveiled general and species specific codon-pair context rules that govern evolution of mRNAs in the 3 domains of life. We show that evolution of bacterial and archeal mRNA primary structure is mainly dependent on constraints imposed by the translational machinery, while in eukaryotes DNA methylation and tri-nucleotide repeats impose strong biases on codon-pair context. CONCLUSIONS: The data highlight fundamental differences between prokaryotic and eukaryotic mRNA decoding rules, which are partially independent of codon usage.

الوصف: Este relatório tem o propósito de refletir criticamente sobre o processo de formação realizado no meu estágio, no ano letivo de 2021/2022, na Escola Secundária do Pinhal Novo. Neste relatório abordo todas as áreas de intervenção que um professor estagiário deverá ter em consideração, tendo em conta o guia de estágio, abrangendo ainda reflexões sobre determinados comportamentos e pensamentos, bem como sobre o processo de formação e ensino aos alunos do 12º ano. Cada área de intervenção é explicada para que o leitor perceba o trabalho realizado durante o ano letivo, querendo com isto que o leitor compreenda a importância de conhecermos o nosso contexto escolar, o porquê de planearmos o nosso ensino, que tipo de estratégias de ensino é que devemos ter em consideração quando damos uma aula, o momento de avaliação e o papel que um professor pode ter na sua escola, pois o foco não é apenas naquilo que é denominado dar aulas. É difícil descrever com exatidão todos os procedimentos realizados para cada uma das áreas, no entanto, procurei analisar e refletir sobre os momentos que considero essenciais durante o meu estágio. De salientar que termino o meu relatório com uma reflexão que poderá servir de exemplo para futuros professores estagiários, ajudando-os a serem melhores profissionais.

الوصف (مترجم): This report aims to critically reflect on the training process carried out during my internship, in the academic year 2021/2022, at Escola Secundária do Pinhal Novo. In this report I address all the areas of intervention that a trainee teacher should take into account, taking into account the internship guide, as reflections on certain behaviors and thoughts, on the training and teaching process for 12th grade students. Each intervention area is explained so that the reader understands the work carried out during the school year, with this wanting the reader to understand the importance of knowing our school context, why we plan our teaching, what kind of teaching strategies are that we should take into account when we teach a class, the moment of evaluation and the role that a teacher can play in his school and not just focus on what it is to teach. It is difficult to accurately describe all the procedures performed for each of the areas, however, I tried to analyze and reflect on moments that I consider essential during my internship. Ending my report with a reflection trying to aim to serve as an example for future trainee teachers helping them to be better professionals.

الوصف: O presente documento visa fazer referência ao estágio que se realizou no ano letivo 2020/2021 na Escola Secundária Morgado de Mateus, com o objetivo de incluir de forma minuciosa todo o trabalho que foi desenvolvido como estagiário na disciplina de Educação Física e, ao mesmo tempo, uma reflexão crítica sobre a experiência pedagógica em contexto escolar. A Prática de Ensino Supervisionada reflete um longo período de experiência e incerteza no sentido em que teremos sempre desafios, dificuldades com as quais não estamos a contar e que acabam por contribuir para o nosso desenvolvimento, onde o estagiário coloca em prática todo o conhecimento adquirido ao longo dos anos e, concomitantemente, valida todas essas competências, obtendo novas aptidões. Vamos ganhando experiência ao longo da Prática de Ensino Supervisionada criando respostas para esses mesmos desafios e conseguindo combater essa incerteza relativamente ao que ainda nos espera acontecer. A presença, a colaboração e a orientação do professor Carlos Pires foi essencial para todo o processo de evolução, uma vez que, desde logo, nos atribuiu durante todo o ano letivo uma turma, diferente por cada período, do ensino secundário que estava sob a nossa responsabilidade. Deste modo, este relatório está estruturado de forma cronológica, do geral para o particular, de forma a abordar todas as partes do processo ensino-aprendizagem. Numa primeira fase será realizada uma breve apresentação em jeito de introdução, seguida de uma exposição de forma a caracterizar as nossas expectativas referentes ao estágio, assim como, o relato da nossa experiência pessoal. Numa segunda fase serão referidos o contexto e o local do estágio, incluindo a caracterização da escola, todos os recursos que a mesma teve à disposição e o departamento de Educação Física. Numa terceira fase será abordada a prática profissional, onde estará incluído todo o processo da vertente prática. Numa quarta e última fase serão abordadas as atividades em que participamos e auxiliámos ao longo do ano escolar. Por fim será realizada uma reflexão crítica acerca de todo o trabalho desenvolvido, assim como, a experiência e aprendizagem adquirida ao longo de todo este processo.

الوصف (مترجم): This document aims to refer to the internship that took place in the school year 2020/2021 at The Morgado Secondary School in Mateus, with the objective of carefully including all the work that was developed as an intern in the discipline of Physical Education and, at the same time, a critical reflection on the pedagogical experience in the school context. The Supervised Teaching Practice reflects a long period of experience and uncertainty, where the trainee puts into practice all the knowledge acquired over the years and, concomitantly, validates all these skills, obtaining new skills. The presence, collaboration and guidance of Professor Carlos Pires was essential for the whole process of evolution, since from the outset he assigned me throughout the school year a different class, for each period, of secondary education that was only my responsibility. Thus, this report is structured chronologically, from general to particular, in order to address all parts of the teaching-learning process. In a first phase, a brief presentation will be held in the form of introduction, followed by an exhibition in order to characterize my expectations regarding the internship, as well as the account of my personal experience. In a second phase will be mentioned the context and place of the internship, including the characterization of the school, all the resources that it has available and the Department of Physical Education. In a third phase, professional practice will be addressed, which will include the whole process of the practical aspect. In a fourth and final phase will be addressed the activities in which I participated and assisted throughout the school year. Finally, a critical reflection will be carried out about all the work developed, as well as the experience and learning acquired throughout this process.

Conseguimos atingir todos os objetivos propostos, tornando-nos assim, Enfermeiros Especialistas no cuidado prestado à pessoa em situação crítica; ABSTRACT: Septic Shock: Nursing Interventions Sepsis and septic shock are one of the biggest health problems, currently being one of the main causes of hospitalization in Intensive Care. Due to its importance, this was the theme we chose to address. This report presents the Major Professional Intervention, characterizing its internship context and reflecting on the entire process of acquiring and developing skills. As a general aim, we defined: to describe and reflect on the activities carried out, correlating the knowledge acquired with clinical practice. Its elaboration was governed by the design methodology, which is the basis of its construction. Through the description and reflection of activities, we were able to substantiate these interventions, articulating them with the existing scientific evidence. We were able to achieve all the proposed objectives, becoming expert nurses in the care to the person in critical situations.

The usage of container-based virtualization technologies keep the multiple environments where a development team works similar enough, that their work is made easier when devel oping and testing new features, which in turn results in a significantly faster delivery. The nature of this tecnhology also brings numerous advantages when it comes to management, monitoring and maintaining resources, allowing for an ease of adjustment, based on the client needs. Throughout this dissertation is presented an extended base of knowledge about container technologies, especially Docker, as well as what are the basic techniques to use when building an application inside such infrastructure, from the writing of the Dockerfile to the adaptation of the multiple pipelines responsible to deploy the application.
Com a rápida evolução da internet nos últimos anos, a prioridade máxima no desenvolvi-mento de software transformou-se de "o quê?" para "quando?". Disponibilizar rapidamente uma aplicação no mercado é agora a vantagem competitiva que todas as empresas ambi-cionam ter. A utilização de tecnologias de virtualização através de containers uniformizam os vários ambientes em que a equipa de desenvolvimento opera, facilitando assim o seu trabalho no que diz respeito à adição e teste de novas funcionalidades, o que resulta numa entrega significativamente mais rápida. A natureza desta tecnologia trás inúmeras vantagens à gestão, monitorização e manutenção de recursos, permitindo facilmente aumentar ou reduzir os mesmos baseado nas necessidades dos seus clientes. Nesta dissertação é apresentada uma extensa base de conhecimento sobre as tecnologias de containerização, em especial o Docker, bem como quais as técnicas base a utilizar quando se pretende construir uma aplicação com uma infraestrutura deste tipo, desde a escrita do Dockerfile, até à adaptação das várias pipelines responsáveis por disponibilizar a aplicação em ambiente de produção.

الوصف: A exploração intensa de fontes de energia não renováveis tem tido um impacto ambiental muito severo. A queima dos combustíveis liberta grandes quantidades de CO2 para a atmosfera, o que contribui para o efeito de estufa provocando um aumento global da temperatura. Neste sentido, é urgente procurar alternativas mais sustentáveis e que, ao mesmo tempo, consigam responder à procura de energia, cada vez mais elevada. En- tre as fontes disponíveis, a energia das ondas apresenta-se como um recurso com um grande potencial. Em Portugal estima-se que a potência presente nas ondas varie entre 8 e 30 kW/m. Atualmente existem diversos conceitos de dispositivos de aproveitamento de ener- gia das ondas (WEC – wave energy converter). No entanto, devido ao facto de ainda não existir uma tecnologia estabelecida, a percentagem de utilização destes WECs é baixa. A divergência na escolha do tipo de WEC pode ser justificada pelas diferentes características geográficas, de agitação ou operacionais. Do ponto de vista operacional, o conversor hiper- bárico de energia de ondas (WEHC - wave energy hyperbaric converter) constitui uma boa alternativa, uma vez que todo o mecanismo de extração de energia encontra-se emerso. O WEHC pode ser facilmente integrado em estruturas costeiras e offshore existentes. Nesta dissertação estuda-se numericamente o comportamento do WEHC em agitação irregular. A modelação é feita usando o código numérico DualSPHysics que se baseia no método SPH (smoothed particle hydrodynamics). Para incluir as restrições mecânicas entre corpos rígidos é utilizada a implementação do ProjectChrono. Todas as simulações foram realizadas com os parâmetros previamente testados por Silva (2019) e Bernardo (2020). Na primeira fase estudou-se o WEHC sem o sistema PTO com o objetivo de averiguar a evolução da resposta do dispositivo. Verificou-se um pico na amplitude para frequências entre 0.15 Hz e 0.17 Hz. Também foram comparadas si- mulações com diferentes alturas e observou-se um aumento do RAO para alturas de onda superiores. Na segunda fase incluiu-se o sistema PTO e foi possível obter informações so- bre a potência e rendimento do WEHC para as diferentes condições de agitação testadas. O CWR (capture width ratio) médio foi de 15%.

الوصف (مترجم): The intensive exploitation of non renewable energy sources has been having a very severe environmental impact. Burning fossil fuels releases large amounts of CO2 into the atmo- sphere which contributes to the greenhouse effect, thus increasing the global temperature. Therefore, it is urgent to seek more sustainable alternatives that, at the same time, can keep up with the increasing energy demand. Among the available sources, wave energy presents itself as a resource with great potential. In Portugal it is estimated that the available wave power varies between 8 and 30 KW/m. Currently, there are several concepts for WECs (Wave Energy Converters). However, due to the fact that there’s no established technology the use percentage of these WECs is very low. The choice of WEC may differ due to different geographic, wave agitation and operational characteristics in the installation sites. Regarding operational characteristics, the WEHC (Wave Energy Hyperbaric Converter) is a good alternative because the energy extraction mechanism is completely emerged. The WEHC can be easily integrated on existing coastal and offshore structures. In this dissertation the WEHC behaviour un- der irregular waves is studied numerically. The modelation is done using the numerical tool DualSPHysics. It uses an SPH (smoothed particle hydrodynamics) method. Pro- jectChrono was used as well to include the mechanical constraints between rigid bodies. All the simulations were run with parameters previously tested by Silva (2019) and Bernardo (2020). In the first stage, the WEHC was studied without the PTO system, in order to evaluate the device’s response. It was observed a peak in amplitude between 0.15 Hz and 0.17 Hz. Simulations with different heights were also compared and it was observed an increase in the RAO parameter for higher wave heights. In the second stage, the PTO system was included and it was possible to obtain information about the absorbed power and efficiency of the device under different wave agitation conditions. The mean CWR (capture width ratio) was 15%.

الوصف: Este trabalho foi realizado no âmbito do estágio curricular para a obtenção do grau de Mestre em Engenharia Mecânica. Foca-se na otimização de processos de montagem de sistemas opto-mecânicos, sistemas esses produzidos pela Leica – Aparelhos Ópticos de Precisão S.A. Inicialmente, o estágio começou por uma fase de adaptação e aprendizagem de conceitos óticos gerais e necessários para quem trabalha no meio opto-mecânico, sendo realizada uma análise teórica sobre aberrações, distância focal e entre outros conceitos óticos. Tendo em conta uma abordagem Lean e de modo a aplicar técnicas de otimização durante a montagem dos produtos, começamos por elaborar e analisar o Value Stream Map da linha de montagem, usando a seguir o diagrama de Ishikawa para recolher as causas de um problema em específico. Também foi usada a metodologia 5´S. Após a recolha dos dados, foram implementadas as melhorias necessárias na linha de montagem, melhorias essas que tinham em vista o conforto e bem-estar dos/as colaboradores/as. Foi também realizada uma intervenção na secção da Ferramentaria para a produção e maquinação de uma peça de cariz pessoal. As propostas de melhoria criaram uma melhor fluidez de informação entre processos, assim como a otimização de etapas, reduzindo-se ainda o tempo de torneamento de lentes da objetiva 29mm, sendo refletidas, em parte na OEE do torno mecânico com a melhoria apresentada pela garra de fixação da objetiva de 29mm devido ao surgimento de deformações nos casquilhos das mesmas. Adicionalmente a estas melhorias na linha de montagem das objetivas SALSA (Prime), foram implementadas também melhorias na restante linha de montagem de produtos externos à “casa mãe”, não sendo estas o principal foco deste trabalho.

الوصف (مترجم): This work was carried out as part of the curricular internship to obtain a Master's degree in Mechanical Engineering. Focusing on optimizing assembly processes for opto-mechanical systems, these systems are produced by Leica – Optical Precision Devices S.A. Initially, the internship began with a phase of adaptation and learning of general optical concepts required and necessary for those working in opto-mechanical environments, with a theoretical analysis of aberrations, focal length and other optical concepts being carried out. Considering a Lean approach and in order to apply optimization techniques during the assembly of the products, we start by elaborating and analysing the Value Stream Map of the assembly line, then using the Ishikawa diagram to collect the causes of a specific problem. The 5'S methodology was also implemented. After collecting the data, the necessary improvements were implemented in the assembly line, improvements aimed at the comfort and well-being of employees. An intervention was also carried out in the Tooling section for the production and machining of a personal part. The improvement proposals created a better fluidity of information between processes, as well as the optimization of steps, also reducing the turning time of 29mm objective lenses, being reflected, in part, in the OEE of the lathe with the improvement presented by the claw 29mm objective attachment. In addition to these improvements in the assembly line for SALSA lenses (Prime), improvements were also implemented in the remaining assembly line for products outside the “mother house”.
