مستخلص: P21 is a protein secreted by all forms of Trypanosoma cruzi (T. cruzi) with recognized biological activities determined in studies using the recombinant form of the protein. In our recent study, we found that the ablation of P21 gene decreased Y strain axenic epimastigotes multiplication and increased intracellular replication of amastigotes in HeLa cells infected with metacyclic trypomastigotes. In the present study, we investigated the effect of P21 in vitro using C2C12 cell lines infected with tissue culture-derived trypomastigotes (TCT) of wild-type and P21 knockout (TcP21-/-) Y strain, and in vivo using an experimental model of T. cruzi infection in BALB/c mice. Our in-vitro results showed a significant decrease in the host cell invasion rate by TcP21-/- parasites as measured by Giemsa staining and cell count in bright light microscope. Quantitative polymerase chain reaction (qPCR) analysis showed that TcP21-/- parasites multiplied intracellularly to a higher extent than the scrambled parasites at 72h post-infection. In addition, we observed a higher egress of TcP21-/- trypomastigotes from C2C12 cells at 144h and 168h post-infection. Mice infected with Y strain TcP21-/- trypomastigotes displayed higher systemic parasitemia, heart tissue parasite burden, and several histopathological alterations in heart tissues compared to control animals infected with scrambled parasites. Therewith, we propose that P21 is important in the host-pathogen interaction during invasion, cell multiplication, and egress, and may be part of the mechanism that controls parasitism and promotes chronic infection without patent systemic parasitemia. الملخص (بالإنجليزية): Introduction: NDM-1-producing is increasingly common in new generation of multidrug-resistant bacteria. Objective and Methods: This study reports the occurrence of metallo-ß-lactamase, especially NDM in Pseudomonas aeruginosa in the state of Minas Gerais, Brazil. A disc combination test used to screen isolates and Polymerase Chain Reaction was used to search for metallo-ß-lactamases genes. Results: Among the 95 strains analyzed, all were resistant to carbapenems, 20% were phenotypically positive for metallo-ß-lactamases, and 1% were positive for blaSPM gene and 2.1% for blaNDM. Conclusions: In Brazil, SPM carbapenemases always was predominant among P. aeruginosa clinical isolates, but not NDM. This study highlights the need to review clinical protocols, in order to contain the dissemination of this profile. [ABSTRACT FROM AUTHOR]
Abstract (Portuguese): Introdução: A produção de NDM-1 é cada vez mais comum nesta nova geração de bactérias multirresistentes. Objetivo e Métodos: Este estudo avalia a ocorrência de metalo-ß-lactamase, especialmente NDM em Pseudomonas aeruginosa no estado de Minas Gerais, Brasil, por meio de um teste do disco combinado para triagem de isolados e Reação em Cadeia de Polimerase para pesquisar os genes para metalo-ß-lactamases. Resultados: Das 95 cepas analisadas, todas resistentes aos carbapenêmicos, 20% eram fenotipicamente positivas para metalo-ß-lactamases, e 1% positivas para o gene blaSPM e 2,1% para blaNDM. Conclusões: No Brasil, as carbapenemases SPM sempre foram predominantes entre os isolados clínicos de P. aeruginosa, mas não a NDM. Este estudo destaca a necessidade de revisão de protocolos clínicos, a fim de conter a disseminação desse novo perfil. [ABSTRACT FROM AUTHOR]

الملخص (بالإنجليزية): Objective: This study aimed to evaluate the quality of life, through the "Medical Outcomes Study 36 - Item Short - Form Health Survey" (SF-36) questionnaire, of a group of elderly involved in physical activities scheduled and the impact of physical activity time. Methods: We assessed 143 elderly engaged in physical activity programmed by Universidade Federal de Uberlândia. As a data collection tool, we used the SF-36 and a sociodemographic questionnaire, applied at the time of the interview. Statistical analysis was performed using Kruskal-Wallis, for analysis between domains, and Mann-Whitney test, to verify the relationship between health status and physical activity level. Results: The average age was 70.5 years. The mean score for the SF-36 of the elderly people who participated in the research was 73.3. The best result was in the Social Aspects domain (81.7), followed by Mental Health (78.9). Approximately 76% had at least 150 minutes of physical activity per week, being classified as a more active population. Most (70.6%) of the elderly had a good perception of their general health. There was statistical difference in the General Health domain among groups that performed physical activity for a period of less than one year and those who have been engaging in regular physical activity for over 10 years. Conclusions: The Social Function and Mental Health domains had the highest scores, with significant documentation of a better general health in the group that have practiced consecutive physical activity for over ten years. Abstract (Portuguese): Objetivos: Avaliar a qualidade de vida, através do questionário "Medical Outcomes Study 36 - Item Short - Form Health Survey" (SF-36) em um grupo de idosos envolvidos em atividades físicas programadas e o impacto do tempo de atividade física. Métodos: Avaliamos 143 idosos envolvidos em atividade física programada pela Universidade Federal de Uberlândia. Como ferramenta de coleta de dados, foram utilizados os questionários SF-36 e um sociodemográfico, aplicados no momento da entrevista. Foi realizado o teste Kruskal-Wallis para análise entre os domínios e Mann-Whitney para verificar a relação entre o estado de saúde e o nível de atividade física. Resultados: A média de idade da população foi de 70,5 anos. A pontuação média encontrada a partir da análise do SF-36 foi de 73,3. Os domínios Aspectos Sociais (81,7) e Saúde Mental (78,9) apresentaram os escores mais elevados. Cerca de 76% dos idosos incluídos no estudo praticavam pelo menos 150 minutos de atividade física por semana, sendo classificada como população mais ativa. A maioria (70,6%) dos idosos tinha uma boa percepção do seu estado geral de saúde; entretanto, observou-se diferença estatística quanto ao domínio Estado Geral de Saúde entre os grupos que praticavam atividade física por período menor que um ano e aqueles que praticam há mais de 10 anos. Conclusões: As melhores pontuações foram obtidas para os domínios Aspectos Sociais e Saúde Mental, sendo observado significativo melhor estado geral de saúde no grupo que pratica atividade física consecutiva há mais de dez anos. الملخص (بالإنجليزية): The rates of multidrug resistant Gram-negative bacilli (MDR-GNB) have risen substantially worldwide, being associated with impact adverse on outcome of patients with severe infections. The aims of the study were to identify the risk factors associated with MDRGNB- related bacteremia, and evaluate patient's outcome. We also assessed the prevalence of resistance phenotypes in GNB isolated from the blood and catheter tip. A case-control study was performed to evaluate risk factors for development of MDR-GNB-related bacteremia at inpatients at a tertiary university hospital from May 2009 through February 2010. A cohort study was conducted for determine the predictors of death. It was considered GNB isolated from all episodes of bacteremia and microorganisms recovered from catheter tip for analysis of the production of Extended-spectrumbeta- lactamase (ESBL) and Metallo-beta-lactamase (MBL). MDRGNB bacteremia occurred in 33.6% of patients and A. baumannii (55%) was the most frequent. The rate of ESBL-producing isolates was 58% and 32% for Klebsiella spp. and E. coli, respectively. The MBL was detected in 34% of the P. aeruginosa isolates. MDRGNB bacteremia was associated extrinsic factors such as: surgery, tracheostomy and inadequate therapy. The risk for death was related to advanced age, MDR-GNB bacteremia, heart disease and inadequate therapy. [ABSTRACT FROM AUTHOR]
Abstract (Portuguese): As taxas de bacilos Gram-negativos multirresistentes (MDR-GNB) têm aumentado substancialmente em todo o mundo, estando associada ao um impacto negativo na evolução dos pacientes com infecções graves. Os objetivos do estudo foram identificar os fatores de risco associados com bacteremia por MDR-GNB e avaliar a evolução do paciente. Também avaliamos a prevalência de fenótipos de resistência em GNB isolados do sangue e ponta do cateter. Um estudo caso-controle foi conduzido para avaliar os fatores de risco associados com bacteremia por MDR-GNB em pacientes internados num hospital universitário terciário, durante o período de maio de 2009 a fevereiro de 2010. Outro estudo de coorte foi realizado para determinar os preditores de mortalidade, no qual foram incluídos todos os micro-organismos isolados do sangue (todos os episódios de bacteremia) e todos aqueles recuperados a partir da ponta do cateter, para análise da produção de Beta-lactamase de amplo espectro (ESBL) e Metalo-beta-lactamase (MBL). Bacteremia por MDR- -GNB ocorreu em 33,6% dos pacientes, e A. baumannii (55%) foi o isolado mais frequente. A taxa de isolados produtores de ESBL foi de 58% e 32% para Klebsiella spp. e E. coli, respectivamente. A produção de MBL foi detectada em 34% dos isolados de P. aeruginosa. Bacteremia por MDR-GNB foi associado com fatores extrínsecos, tais como: cirurgia, traqueostomia e terapia inadequada. O risco de morte foi relacionado com a idade avançada, bacteremia por MDR-GNB, doença cardíaca e terapia antimicrobiana inadequada. [ABSTRACT FROM AUTHOR]
