    الوصف: We address the problem of synthesizing safety-critical cyber-physical system architectures to minimize a cost function while guaranteeing the desired reliability. We cast the problem as an integer linear program on a reconfigurable graph which models the architecture. Since generating symbolic probability constraints by exhaustive enumeration of failure cases on all possible graph configurations takes exponential time, we propose two algorithms to decrease the problem complexity, i.e. Integer-Linear Programming Modulo Reliability (ILP-MR) and Integer-Linear Programming with Approximate Reliability (ILP-AR). We compare the two approaches and demonstrate their effectiveness on the design of aircraft electric power system architectures.

    الوصف: We use a multiscale approach to study a lattice-gas model of submonolayer growth of Fe/Mo(110) by Molecular Beam Epitaxy. To begin with, we construct a two-dimensional lattice-gas model of the Fe/Mo(110) system based on first-principles calculations of the monomer diffusion barrier and adatom-adatom interactions. The model is investigated by equilibrium Monte Carlo (MC) simulations to compute the diffusion coefficients of Fe islands of different sizes. These quantities are then used as input to the coarse-grained Kinetic Rate Equation (KRE) approach, which provides time evolution of the island size distributions for a system undergoing diffusion driven aggregation within the 2D submonolayer regime. We calculate these distributions at temperatures T=500 K and 1000 K using the KRE method. We also employ direct kinetic MC simulations of our model to study island growth at T=500 K, and find good agreement with the KRE results, which validates our multi-scale approach.

    الوصف: Deep learning (DL) algorithms have already proved their effectiveness on a wide variety of application domains, including speech recognition, natural language processing, and image classification. To foster their pervasive adoption in applications where low latency, privacy issues and data bandwidth are paramount, the current trend is to perform inference tasks at the edge. This requires deployment of DL algorithms on low-energy and resource-constrained computing nodes, often heterogenous and parallel, that are usually more complex to program and to manage without adequate support and experience. In this paper, we present ALOHA, an integrated tool flow that tries to facilitate the design of DL applications and their porting on embedded heterogenous architectures. The proposed tool flow aims at automating different design steps and reducing development costs. ALOHA considers hardware-related variables and security, power efficiency, and adaptivity aspects during the whole development process, from pre-training hyperparameter optimization and algorithm configuration to deployment.

    الوصف: Introduction: This study aimed to address therapeutic inertia in the management of type 2 diabetes (T2D) by investigating the potential of early treatment with oral semaglutide. Methods: A cross-sectional survey was conducted between October 2021 and April 2022 among specialists treating individuals with T2D. A scientific committee designed a data collection form covering demographics, cardiovascular risk, glucose control metrics, ongoing therapies, and physician judgments on treatment appropriateness. Participants completed anonymous patient questionnaires reflecting routine clinical encounters. The preferred therapeutic regimen for each patient was also identified. Results: The analysis was conducted on 4449 patients initiating oral semaglutide. The population had a relatively short disease duration (42% < 5 years), and a minority (15.6%) had a history of cardiovascular events. Importantly, oral semaglutide was started in subjects with various disease durations and background therapies. Notably, its initiation was accompanied by de-prescription of sulfonylureas, pioglitazone, DPP-4 inhibitors, and insulin. Choice of oral semaglutide was influenced by patient profiles and ongoing glucose-lowering regimens. Factors such as younger age, higher HbA1c, and ongoing SGLT-2 inhibitor therapy drove the choice of oral semaglutide with the aim of improving glycemic control. Projected glycemic effectiveness analysis revealed that oral semaglutide could potentially lead HbA1c to target in > 60% of patients, and more often than sitagliptin or empagliflozin. Conclusion: The study supports the potential of early implementation of oral semaglutide as a strategy to overcome therapeutic inertia and enhance T2D management.

    الوصف: The use of Deep Learning (DL) algorithms is increasingly evolving in many application domains. Despite the rapid growing of algorithm size and complexity, performing DL inference at the edge is becoming a clear trend to cope with low latency, privacy and bandwidth constraints. Nevertheless, traditional implementation on low-energy computing nodes often requires experience-based manual intervention and trial-and-error iterations to get to a functional and effective solution. This work presents a computer-aided design (CAD) support for effective implementation of DL algorithms on embedded systems, aiming at automating different design steps and reducing cost. The proposed tool flow comprises capabilities to consider architecture-and hardware-related variables at very early stages of the development process, from pre-training hyperparameter optimization and algorithm configuration to deployment, and to adequately address security, power efficiency and adaptivity requirements. This paper also presents some preliminary results obtained by the first implementation of the optimization techniques supported by the tool flow.

The goal of Technical Requirements Elicitation (TRE) is to ensure that all needs of involved stakeholders are being identified and adequately addressed without prescribing how to achieve them. Whilst TRE methodology in product or service development is well known, TRE in large research projects turns far too commonly into an ad-hoc process carried out without the support of a common, solid methodology. The objective of this paper is to propose a methodology for identification of all stakeholders in research projects and their needs based on experience of Horizon 2020 project CERBERO.

    الوصف: The highly pathogenic Old World arenavirus Lassa virus (LASV) and the prototypic arenavirus lymphocytic choriomeningitis virus (LCMV) use α-dystroglycan as a cellular receptor and enter the host cell by an unusual endocytotic pathway independent of clathrin, caveolin, dynamin, and actin. Upon internalization, the viruses are delivered to acidified endosomes in a Rab5-independent manner bypassing classical routes of incoming vesicular trafficking. Here we sought to identify cellular factors involved in the unusual and largely unknown entry pathway of LASV and LCMV. Cell entry of LASV and LCMV required microtubular transport to late endosomes, consistent with the low fusion pH of the viral envelope glycoproteins. Productive infection with recombinant LCMV expressing LASV envelope glycoprotein (rLCMV-LASVGP) and LCMV depended on phosphatidyl inositol 3-kinase (PI3K) as well as lysobisphosphatidic acid (LBPA), an unusual phospholipid that is involved in the formation of intraluminal vesicles (ILV) of the multivesicular body (MVB) of the late endosome. We provide evidence for a role of the endosomal sorting complex required for transport (ESCRT) in LASV and LCMV cell entry, in particular the ESCRT components Hrs, Tsg101, Vps22, and Vps24, as well as the ESCRT-associated ATPase Vps4 involved in fission of ILV. Productive infection with rLCMV-LASVGP and LCMV also critically depended on the ESCRT-associated protein Alix, which is implicated in membrane dynamics of the MVB/late endosomes. Our study identifies crucial cellular factors implicated in Old World arenavirus cell entry and indicates that LASV and LCMV invade the host cell passing via the MVB/late endosome. Our data further suggest that the virus-receptor complexes undergo sorting into ILV of the MVB mediated by the ESCRT, possibly using a pathway that may be linked to the cellular trafficking and degradation of the cellular receptor.

    الوصف: BACKGROUND: Alterations in glucose metabolism and epithelial-mesenchymal transition (EMT) constitute two important characteristics of carcinoma progression toward invasive cancer. Despite an extensive characterization of each of them separately, the links between EMT and glucose metabolism of tumor cells remain elusive. Here we show that the neuronal glucose transporter GLUT3 contributes to glucose uptake and proliferation of lung tumor cells that have undergone an EMT. RESULTS: Using a panel of human non-small cell lung cancer (NSCLC) cell lines, we demonstrate that GLUT3 is strongly expressed in mesenchymal, but not epithelial cells, a finding corroborated in hepatoma cells. Furthermore, we identify that ZEB1 binds to the GLUT3 gene to activate transcription. Importantly, inhibiting GLUT3 expression reduces glucose import and the proliferation of mesenchymal lung tumor cells, whereas ectopic expression in epithelial cells sustains proliferation in low glucose. Using a large microarray data collection of human NSCLCs, we determine that GLUT3 expression correlates with EMT markers and is prognostic of poor overall survival. CONCLUSIONS: Altogether, our results reveal that GLUT3 is a transcriptional target of ZEB1 and that this glucose transporter plays an important role in lung cancer, when tumor cells loose their epithelial characteristics to become more invasive. Moreover, these findings emphasize the development of GLUT3 inhibitory drugs as a targeted therapy for the treatment of patients with poorly differentiated tumors.
