    الوصف: Rogaland in southwest Norway was a core production area for bucket-shaped pottery throughout the ca. 200-year period spanned by these finds. Largely thanks to Elna Siv Kristoffersen's work we have a well-developed understanding of the final century of this characteristic Migration Period find: certain ceramic craft networks rose to prominence, culminating in workshop milieux intimately tied to the formation of central places like those in Jæren, Rogaland from around AD 450/60, eventually making bucket-shaped pots alongside Style I metalwork. This inventive cross-craft focus notwithstanding, we know less about the first century of production. A recent study suggests that the rise of the Jæren workshop milieux was concurrent with a gradual decline of the Augland ceramic workshop, related to the Oddernes elite milieu in Vest-Agder. Consequently, the areas around and between these two regional nodal points have come to be of particular interest. What happened to connectivity in this hinterland during the emergent first century of bucket-shaped ceramic production? This batch study identifies paste recipes and traces the movements of pots. Cognisant of the lack of comprehensive archaeometric studies, partly due to costs, we present a transferrable and relatively inexpensive approach that combines qualitative macroscopy with quantitative analysis of data from a handheld X-ray fluorescence (h-XRF) device

    الوصف: The discussion of the transition to farming in southern Africa and the formation of Early Iron Age society, referred to in Mozambican archaeology as the Early Farming Communities (EFC), is complicated by the lack of surveys in key areas of intensive contacts. This article presents the results of excavations at the EFC site Chicumbane in the eastern lower Limpopo Valley, dated 500–800 AD. The variation of ceramic styles in terms of decoration and shape suggests predominantly interior influences (Gokomere and Zhizo facies), but there are also decoration elements similar to what is found on the coast. A wide variety of ceramic technologies were used in terms of clay sources, temper, and ways of building the pot. Here, we reconstruct possible social interactions based on these differences. Together with the other artifact categories, such as slag, metal, and shell beads, the results show some aspects of regional interactions among Early Farming Communities. The combined ceramic analyses suggest a mix of traditions by female potters who, through marriage, moved between regions, bringing new ways of decorating, tempering, and building pots.

    الوصف: Long-term stability and biological safety are crucial for translation of 3D-bioprinting technology into clinical applications. Here, we addressed the long-term safety and stability issues associated with 3D-bioprinted constructs comprising a cellulose scaffold and human cells (chondrocytes and stem cells) over a period of 10 months in nude mice. Our findings showed that increasing unconfined compression strength over time significantly improved the mechanical stability of the cell-containing constructs relative to cell-free scaffolds. Additionally, the cell-free constructs exhibited a mean compressive stress and stiffness (compressive modulus) of 0.04 +/- 0.05 MPa and 0.14 +/- 0.18 MPa, respectively, whereas these values for the cell-containing constructs were 0.11 +/- 0.08 MPa (p= .019) and 0.53 +/- 0.59 MPa (p= .012), respectively. Moreover, histomorphologic analysis revealed that cartilage formed from the cell-containing constructs harbored an abundance of proliferating chondrocytes in clusters, and after 10 months, resembled native cartilage. Furthermore, extension of the experiment over the complete lifecycle of the animal model revealed no signs of ossification, fibrosis, necrosis, or implant-related tumor development in the 3D-bioprinted constructs. These findings confirm the in vivo biological safety and mechanical stability of 3D-bioprinted cartilaginous tissues and support their potential translation into clinical applications.

    الوصف: BACKGROUND: Since the warnings by the United States (US) and European regulatory authorities in 2004 and 2005 it had been discussed whether there is some link between selective serotonin reuptake inhibitors (SSRIs) and suicidality in the pediatric population. The aim of our study was to describe trends and patterns in spontaneous reporting data referring to suicidality in children, adolescents and young adults treated with SSRI after the warnings. METHODS: Descriptive analyses of reports for 0–24 year olds referring to suicide/suicidal ideations, self-harms and overdoses with SSRIs reported as suspected submitted to the US (FAERS) and the European (EudraVigilance) adverse drug reaction databases until 2019 were performed. The causal relationship was assessed in accordance with the WHO criteria for the European reports. For Germany, prescription data for SSRIs were provided and reporting rates (number of reports/number of prescriptions) were calculated for the reports with possible causal relationship (so called “confirmed reports”). RESULTS: Since 2004, the number of reports referring to suicide/suicidal ideations, self-harm and overdoses increased steadily in the US and EU. However, only a slight increase was seen for the confirmed EU reports. After 2008, the proportion of reports informing about suicidal ideations increased, while the proportion of fatal suicide attempts decreased. Reporting rates were higher for females and adolescents (12-18 years). CONCLUSIONS: Our results demonstrate the importance of further monitoring suicidality in 0–24 year olds treated with SSRI in order to recognize suicidality early avoiding fatal suicide attempts. The higher reporting rates for females and adolescents should be further investigated.

    الوصف: Among Simon Hall's influential contributions to historical archaeology are two research agendas: The need to focus attention on lower scalar levels of analysis, and broadening the concept of ceramic style to include less visible technological qualities. The latter is of particular importance to the stylistically bland and less decorated assemblages from the 18th and 19th centuries. Combining and developing the two agendas further, this article presents a new set of analyses of ceramic material from the stonewalled sites Marothodi and Lebenya in the Magaliesberg region, dating to the decades leading up to the difaqane in the 1820s. We explore households as flexible spaces for making, creativity and memory-work in turbulent times. The late 18th and early 19th centuries saw an accelerated development of pyrotechnologies such as metalworking and ceramics. This happened in tandem with significant changes to the built environment and spatial organisation of the household, which was the primary arena for craft learning. Frequent relocation and alteration of learning spaces put transmission and teacher apprentice ties under serious strain. Seeking to trace connections across a complex and layered political landscape, we tentatively hypothesise that ceramic craftspeople became relatively less reliant on locally anchored insights and placed more emphasis on sharing knowledge and materials within extended craft-learning networks. The study includes a comparison of the results of petrographic and geochemical laboratory analyses with those from a handheld XRF device. Offering instant feedback while still in the field, such mobile tools can help in developing sampling strategies that also include a higher percentage of undecorated ceramic material.

    الوصف: The different chambers of the human heart demonstrate regional physiological traits and may be differentially affected during pathologic remodeling, resulting in heart failure. Few previous studies have, however, characterized the different chambers at a transcriptomic level. We therefore conducted whole-tissue RNA sequencing and gene set enrichment analysis of biopsies collected from the four chambers of adult failing (n = 8) and nonfailing (n = 11) human hearts. Atria and ventricles demonstrated distinct transcriptional patterns. Compared to nonfailing ventricles, the transcriptional pattern of nonfailing atria was enriched for a large number of gene sets associated with cardiogenesis, the immune system and bone morphogenetic protein (BMP), transforming growth factor beta (TGF beta), MAPK/JNK and Wnt signaling. Differences between failing and nonfailing hearts were also determined. The transcriptional pattern of failing atria was distinct compared to that of nonfailing atria and enriched for gene sets associated with the innate and adaptive immune system, TGF beta/SMAD signaling, and changes in endothelial, smooth muscle cell and cardiomyocyte physiology. Failing ventricles were also enriched for gene sets associated with the immune system. Based on the transcriptomic patterns, upstream regulators associated with heart failure were identified. These included many immune response factors predicted to be similarly activated for all chambers of failing hearts. In summary, the heart chambers demonstrate distinct transcriptional patterns that differ between failing and nonfailing hearts. Immune system signaling may be a hallmark of all four heart chambers in failing hearts, and could constitute a novel therapeutic target.

    الوصف: Background: Autologous conditioned serum (ACS) is used to treat osteoarthritis in horses, although its effects are not fully investigated. Objectives: To investigate the effects of equine serum and conditioned serum on chondrocytes stimulated with interleukin (IL)-1 beta and cartilage explants with mild osteoarthritis. Study design: In vitro experimental study. Methods: The effect of three different serum preparations (unincubated control [PS], serum incubated 24 h [PS24h] and serum incubated 24 h in ACS containers [PCS]) pooled from lame horses were tested in two in vitro models. IL-1 beta and IL-1 receptor antagonist (IL-1Ra) concentrations were measured in all sera. In model 1, chondrocyte pellet cultures were stimulated with IL-1 beta prior to treatment with the serum preparations for 2 and 48 h. Microarray, polymerase chain reaction, and matrix metallopeptidase-13 analyses were performed. In model 2, cartilage explants from horses with structural osteoarthritis were treated with PS or PCS on days 0, 6 and 12, or left untreated, and evaluated at day 24 using the OARSI grading scale for histological evaluation of articular cartilage. Results: The IL-1Ra concentration in PS24h and PCS was significantly higher than in PS. In model 1, inflammation- and cartilage matrix degradation-related genes were upregulated after 48 h in all treatment groups versus untreated controls. Cartilage matrix molecules, aggrecan and collagens, were downregulated in PS24h- and PCS-treated pellets versus untreated controls. Growth factor signalling genes were upregulated-FGF7 in all treatment groups, BMP2 in PS24h-, and INHBA in PCS-treated-compared with untreated controls. In model 2, the OARSI score at day 24 was not significantly different between treatment groups. Main limitations: Results from in vitro models cannot be directly translated to in vivo situations. Conclusions: In vitro treatment with conditioned serum did not alleviate IL-1 beta-induced responses in chondrocyte pellets or lead to morphological improvement in osteoarthritic cartilage explants.
