الوصف: Background N6-Methyladenosine (m6A) methylation is the most prevalent post-transcriptional modification in mRNA, and plays significant roles in various diseases. Nevertheless, the precise functions of m6A modification in the formation of ALI remain unclear. In this study we explore the transcriptome distribution of m6A methylation and its probable roles of in ALI. Methods Lipopolysaccharide (LPS) was utilized to establish an ALI mouse model. Real-time qPCR, Western blotting and m6A dot blot were utilized to assess m6A methylation level and the expression of m6A methylation enzymes. MeRIP-Seq and RNA-seq were utilized to explore differential m6A modifications and differentially expressed genes in ALI mice. The hub genes and enriched pathways were assessed by Real-time qPCR and Western blotting. Results Our findings showed that overall m6A methylation level was increased in ALI mice lung tissues, accompanied by lower levels of METTL3 and FTO. Notably, the protein expression of these methylases were different in various cells. There were 772 differently expressed m6A peaks in ALI as compared to the control group, with 316 being hypermethylated and 456 being hypomethylated. GO and KEGG analyses demonstrated these differentially methylated genes were associated with the calcium signaling pathway and cAMP signaling pathway. Furthermore, we identified 50 genes with distinct m6A peaks and mRNA expressions by combined analysis of MeRIP-Seq and RNA-Seq. KEGG analysis also demonstrated that these overlapped genes were closely associated with the calcium signaling pathway, cGMP-PKG signaling pathway, etc. Besides, Western blotting results demonstrated that the protein expression of Fibronectin leucine-rich transmembrane protein 3 (Flrt3) as well as the calcium signaling pathway and cGMP-PKG signaling pathway, increased significantly after ALI. Conclusions m6A modification was paramount in the pathogenesis of ALI, and provided a foundation for the further investigation in the prevention and treatment of ALI.

الوصف: The pollution of soil with heavy metals is an increasingly serious worldwide problem, and cadmium (Cd) has attracted attention because of its high toxicity to almost all plants. Since castor tolerates the accumulation of heavy metals, it has the potential for heavy metal soil remediation. We studied the mechanism of the tolerance of castor to Cd stress treatments at three doses: 300 mg/L, 700 mg/L, and 1,000 mg/L. This research provides new ideas for revealing the defense and detoxification mechanisms of Cd-stressed castor. By combining the results of physiology, differential proteomics and comparative metabolomics, we conducted a comprehensive analysis of the networks that regulate the response of castor to Cd stress. The physiological results mainly emphasize the super-sensitive responses of castor plant roots to Cd stress and the effects of Cd stress on plants’ antioxidant system, ATP synthesis and ion homeostasis. We confirmed these results at the protein and metabolite levels. In addition, proteomics and metabolomics indicated that under Cd stress, the expressions of proteins involved in defense and detoxification, energy metabolism and other metabolites such as organic acids and flavonoids were significantly up-regulated. At the same time, proteomics and metabolomics also show that castor plants mainly block the root system’s absorption of Cd2+ by enhancing the strength of the cell wall, and inducing programmed cell death in response to the three different doses of Cd stress. In addition, the plasma membrane ATPase encoding gene (RcHA4), which was significantly upregulated in our differential proteomics and RT-qPCR studies, was transgenically overexpressed in wild type Arabidopsis thaliana for functional verification. The results indicated that this gene plays an important role in improving plant Cd tolerance.

الوصف: Inspired by natural swarms, swarm robotics systems are used in safety-critical tasks due to their scalability, redundancy, and adaptability. However, their design exposes them to two primary vulnerabilities. First, their homogeneity makes them vulnerable to large-scale attacks. Second, logical flaws within swarm algorithms can be exploited, leading to mission failures or crashes. While existing studies can effectively identify these vulnerabilities using system testing and verification, they are often time-consuming and might require repetition following software updates. To this end, we propose a complementary, two-level diversification approach. The first level tackles system homogeneity through software diversification. The second level introduces algorithmic randomness to minimize the exploitability of logical flaws. By leveraging a social force model, we can ensure that the introduced randomized behaviors do not compromise safety. Our evaluations show that the performance overheads remain within acceptable limits, notably at 2% for missions characterized by self-organizing behaviors. ; Published version

الوصف: The existence of causal relationship between dietary factors and respiratory diseases is uncertain. We comprehensively investigated the association between dietary factors and respiratory diseases by using Mendelian randomization (MR). Genetic variants linked to dietary factors were selected as instrumental variables with genome-wide significance. These instrumental variables were obtained from large GWAS databases. These databases include Biobank, the FinnGen study, and other large consortia. We used multivariate MR analyses to control the effects of smoking and education. Median analysis was conducted to evaluate whether body mass index (BMI) played a role in dietary factors in respiratory diseases. Dried fruit intake was found to be associated with a decreased risk of chronic obstructive pulmonary disease (COPD) (OR: 0.211; 95% CI 0.117–0.378; P < 0.001) and asthma (OR: 0.539; 95% CI 0.357–0.815; P = 0.003). Conversely, pork intake was associated with an increased risk of international pharmaceutical federation (IPF) (OR: 1.051*10 2 , 95% CI 4.354–2.56*10 3 , P = 0.004). However, no significant associations were observed between the 20 dietary factors and obstructive sleep apnea (OSA). In addition, multivariate MR analyses showed that the above results were unchanged in smoking and nonsmoking populations, while the effect of dried fruit intake on asthma was significantly attenuated after corrective education. The results of the mediator variable analysis indicated that BMI could serve as a mediator of the above results. This study found that dried fruits slowed the progression of COPD and asthma, while pork promoted IPF. However, no effect of dietary factors on OSA was found. Meanwhile, we showed that the above results were unchanged in smoking and non-smoking populations. In contrast, education could influence the role of diet on asthma, and BMI could be used as a mediating variable to influence the above results.

الوصف: Introduction Conflict monitoring and processing is an important part of the human cognitive system, it plays a key role in many studies of cognitive disorders. Methods Based on a Chinese word-color match Stroop task, which included incongruent and neutral stimuli, the Electroencephalogram (EEG) and functional Near-infrared Spectroscopy (fNIRS) signals were recorded simultaneously. The Pearson correlation coefficient matrix was calculated to analyze brain connectivity based on EEG signals. Granger Causality (GC) method was employed to analyze the effective connectivity of bilateral frontal lobes. Wavelet Transform Coherence (WTC) was used to analyze the functional connectivity of the bilateral hemisphere and ipsilateral hemisphere. Results Results indicated that brain connectivity analysis on EEG signals did not show any significant lateralization, while fNIRS analysis results showed the frontal lobes especially the left frontal lobe play the leading role in dealing with conflict tasks. The human brain shows leftward lateralization while processing the more complicated incongruent stimuli. This is demonstrated by the higher functional connectivity in the left frontal lobe and the information flow from the left frontal lobe to the right frontal lobe. Discussion Our findings in brain connectivity during cognitive conflict processing demonstrated that the dual modality method combining EEG and fNIRS is a valuable tool to excavate more information through cognitive and physiological studies.
