    الوصف: IntroductionSorafenib is currently the first-line treatment for patients with advanced hepatocellular carcinoma (HCC). Nevertheless, sorafenib resistance remains a huge challenge in the clinic. Therefore, it is urgent to elucidate the mechanisms underlying sorafenib resistance for developing novel treatment strategies for advanced HCC. In this study, we aimed to investigate the role and mechanisms of interleukin-22 (IL-22) in sorafenib resistance in HCC.MethodsThe in vitro experiments using HCC cell lines and in vivo studies with a nude mouse model were used. Calcium staining, chromatin immunoprecipitation, lactate dehydrogenase release and luciferase reporter assays were employed to explore the expression and roles of IL-22, STAT3 and CD155 in sorafenib resistance.ResultsOur clinical results demonstrated a significant correlation between elevated IL-22 expression and poor prognosis in HCC. Analysis of transcriptomic data from the phase-3 STORM-trial (BIOSTORM) suggested that STAT3 signaling activation and natural killer (NK) cell infiltration may associate sorafenib responses. STAT3 signaling could be activated by IL-22 administration in HCC cells, and then enhanced sorafenib resistance in HCC cells by promoting cell proliferation and reducing apoptosis in vitro and in vivo. Further, we found IL-22/STAT3 axis can transcriptionally upregulate CD155 expression in HCC cells, which could significantly reduce NK cell-mediated HCC cell lysis in a co-culture system.ConclusionsCollectively, IL-22 could contribute to sorafenib resistance in HCC by activating STAT3/CD155 signaling axis to decrease the sensitivities of tumor cells to sorafenib-mediated direct cytotoxicity and NK cell-mediated lysis. These findings deepen the understanding of how sorafenib resistance develops in HCC in terms of IL-22/STAT3 signaling pathway, and provide potential targets to overcome sorafenib resistance in patients with advanced HCC.

    الوصف: Background. Lamin family members play crucial roles in promoting oncogenesis and cancer development. The values of lamin family in predicting prognosis and immunotherapy response remain largely unclarified. Our research is aimed at comprehensively estimating the clinical significance of lamin family in hepatocellular carcinoma and constructing a novel lamin family-based signature to predict prognosis and guide the precise immunotherapy. Methods. The expression features and prognostic value of LMNA, LMNB1, and LMNB2 were explored in the TCGA and GEO databases. The biological functions of LMNB1 and LMNB2 were validated by in vitro assays. A lamin family-based signature was built using the TCGA training set. The TCGA test set, entire TCGA set, and GSE14520 set were used to validate its predictive power. Univariate and multivariate analyses were performed to evaluate the independence of the lamin family-based signature from other clinicopathological characteristics. A nomogram was constructed using the lamin family-based signature and TNM stage. The associations of this signature with molecular pathways, clinical characteristics, immune cell infiltration, and immunotherapy response were analyzed. Results. Lamin family members were upregulated in HCC. Upregulation of LMNB1 and LMNB2 promoted HCC proliferation, migration, and invasion. The predictive signature was initially established based on LMNB1 and LMNB2 which could effectively identify differences in overall survival, immune cell infiltration, and clinicopathological characteristics of high- and low-risk patients. The nomogram showed high prognostic predictive accuracy. Importantly, the lamin family-based signature was correlated with immune suppression and expression of immune checkpoint molecules. Conclusions. The lamin family-based signature is a robust biomarker to predict overall survival and immunotherapy response in HCC. High-risk score patients have a poorer overall survival and might be more sensitive to immunotherapy. This signature may contribute to improving individualized prognosis prediction and precision immunotherapy for HCC patients.

    الوصف: Interest point detection has been widely used in image analysis applications. However, some interest points, including small structures and large angle corners, could not be effectively extracted. This paper proposes a limiting form of median Laplacian of Gaussian (LMLG) filter, which combines the superiority of the traditional Laplacian of Gaussian (LoG) filter and a limiting form of the weighted median LoG filter. A detector is also proposed based on the LMLG filter. The LMLG filter aims to improve the detection of LoG-based methods for interest points, especially small structures and large angle corners. Also, it could detect blobs, edges, and local structures. We conduct the repeatability and discrimination experiments on the Oxford dataset. Moreover, we conduct the recall rate experiment on the DTU dataset. The experiments show that the proposed method achieves comparable performance with state-of-the-art methods. In order to verify the utility of the LMLG detector, we carry out a series of interest point detector-based applications: face recognition, infrared-visible image registration, and image classification. The results demonstrate that the LMLG detector performs better than the nine detectors in face recognition. The LMLG detector outperforms the nine detectors and Hrkać's, Han's and Liu's methods in infrared-visible image registration. Our method also gives a comparable result on image classification.     الوصف: This study takes Cuiwei village, located in Qianshan District of Zhuhai City, as an example. First, it analyzes the human flow activities and activity size of village road a, the visual agglomeration degree and the spatial characteristics of historical sections. Secondly, select the most representative traditional buildings in the village, namely Sanwang temple and Wechsler mansion. When r=n and radius r=3, observe the changes of two parameters of visual integration and visual depth of the two historical buildings. The results show that at this stage, the coordination of villages is poor, people's recognition in the village space is weak, people can't use local perception to reflect the overall space, there is an obvious interface between the inside and outside of the village, the isolation state is obvious, the local intelligibility is good, but the global intelligibility is poor.

    الوصف: With the rapid development of China's economy, the urbanization process is gradually accelerating, and there are a large number of traditional village spaces are abandoned and compressed, which has greatly changed the original pattern of ancient villages. Cuiwei village is an example of Zhuhai's typical Lingnan Cantonese traditional village evolving into a village in the city, which has experienced many changes in spatial pattern. This paper adopts the method of qualitative investigation to conduct a detailed investigation and analysis of the types of historical buildings in Cuiwei village, and focuses on the impact of historical buildings on people's spatial feelings. It puts forward planning suggestions to provide reference for the follow-up protection of the architectural heritage of Cantonese traditional villages.
