    الوصف: This paper presents a hierarchical planning algorithm for racing with multiple opponents. The two-stage approach consists of a high-level behavioral planning step and a low-level optimization step. By combining discrete and continuous planning methods, our algorithm encourages global time optimality without being limited by coarse discretization. In the behavioral planning step, the fastest behavior is determined with a low-resolution spatio-temporal visibility graph. Based on the selected behavior, we calculate maneuver envelopes that are subsequently applied as constraints in a time-optimal control problem. The performance of our method is comparable to a parallel approach that selects the fastest trajectory from multiple optimizations with different behavior classes. However, our algorithm can be executed on a single core. This significantly reduces computational requirements, especially when multiple opponents are involved. Therefore, the proposed method is an efficient and practical solution for real-time multi-vehicle racing scenarios.

    الوصف: Recently, high-order sideband polarimetry has been established as an experimental method that links the polarization of sidebands to an interference of Bloch wavefunctions. However, the robustness of sideband polarizations to increasing dephasing remains to be explored. Here, we investigate the dependence of high-order sideband generation in bulk gallium arsenide on dephasing by tuning temperature. We find that the intensities of the sidebands, but not their polarizations, depend strongly on temperature. Using our polarimetry method, we are able to isolate the contributions of electron-heavy hole (HH) and electron-light hole (LH) pairs to sideband intensities, and separately extract the nonequilibrium dephasing coefficients associated with the longitudinal optical (LO) phonons and acoustic (A) phonons for each species of electron-hole pair. We find that $\Gamma_{\text{HH},\text{A}} = 6.1 \pm 1.6$ $\mu$eV/K, $\Gamma_{\text{LH},\text{A}} < 1.5$ $\mu$eV/K, $\Gamma_{\text{HH},\text{LO}} = 14 \pm 3$ meV, and $\Gamma_{\text{LH},\text{LO}} = 30 \pm 3$ meV.

    الوصف: Abstract Islands and small island groups are among the areas most affected by global climate change and face major adaptation challenges. Their role as hotspots of climate change impacts is widely recognized in the scientific literature, and it is not uncommon for small islands to act as poster children for particular climate-related vulnerability. Nonetheless, many island communities have a long tradition of living with oceanic influences, and the question is whether such habitats, where terrestrial and marine ecosystems overlap, have characteristics that contribute to greater self-regulation and resilience. The question is further whether and how the future or possible futures of such an exposed region and society could unfold, considering the negative impact of climate change and the resulting burden of natural disasters. The Halligen in the southern North Sea are an exceptional example of such a situation, and they are to be examined in this study particularly due to their extremely uncertain future. Questions regarding the future arise even more urgently due to the rapidly advancing sea-level rise in the course of climate change. They were already evident in a local field research in 2014 and for current reasons the collected interview material shall be analyzed under this prospective point of view. So what, under these harsh circumstances, might the futures of the Halligen look like? What does it mean for the future of a habitat and a society when the constant risk of storm surges is multiplied by the global risk of climate change and rising sea level? By examining this question from an interdisciplinary perspective in the present study, multiple answers can be provided. The futures differ significantly depending on the perspective from which one looks into the future. However, it is precisely this multidimensionality and heterogeneity that can pose an advantage when considering the unpredictability of future developments. Choosing this very specific region for the study also adds to the benefit – because: If the conditions change, if the strength of the environmental influences change in regions where two ecosystems overlap and which are particularly vulnerable due to their islandness, dealing with and adapting to the consequences of global warming becomes a glimpse into the future of all of us.

    الوصف: Abstract Extracellular vesicles (EVs) are released by all cells, can cross the blood–brain barrier, and have been shown to play an important role in cellular communication, substance shuttling, and immune modulation. In recent years EVs have shifted into focus in multiple sclerosis (MS) research as potential plasma biomarkers and therapeutic vehicles. Yet little is known about the disease-associated changes in EVs in the central nervous system (CNS). To address this gap, we characterized the physical and proteomic changes of mouse spinal cord-derived EVs before and at 16 and 25 days after the induction of experimental autoimmune encephalomyelitis (EAE), a neuroinflammatory model of MS. Using various bioinformatic tools, we found changes in inflammatory, glial, and synaptic proteins and pathways, as well as a shift in the predicted contribution of immune and glial cell types over time. These results show that EVs provide snapshots of crucial disease processes such as CNS-compartmentalized inflammation, re/de-myelination, and synaptic pathology, and might also mediate these processes. Additionally, inflammatory plasma EV biomarkers previously identified in people with MS were also altered in EAE spinal cord EVs, suggesting commonalities of EV-related pathological processes during EAE and MS and overlap of EV proteomic changes between CNS and circulating EVs.

    الوصف: Advanced in situ techniques based on electrons and X-rays are increasingly used to gain insights into fundamental materials dynamics in liquid media. Yet, ionizing radiation changes the solution chemistry. In this work, we show that ionizing radiation decouples the acidity from autoprotolysis. Consequently, pH is insufficient to capture the acidity of water-based systems under irradiation. Via radiolysis simulations, we provide a more conclusive description of the acid-base interplay. Finally, we demonstrate that acidity can be tailored by adjusting the dose rate and adding pH-irrelevant species. This opens up a huge parameter landscape for studies involving ionizing radiation.

    الوصف: Abstract Complementary to synaptophysin and chromogranin A, insulinoma‐associated protein 1 (INSM1) has emerged as a sensitive marker for the diagnosis of neuroendocrine neoplasms. Since there are no comparative data regarding INSM1 expression in conventional colorectal adenocarcinomas (CRCs) and colorectal mixed adenoneuroendocrine carcinomas/neuroendocrine carcinomas (MANECs/NECs), we examined INSM1 in a large cohort of conventional CRCs and MANECs/NECs. In conventional CRC, we put a special focus on conventional CRC with diffuse expression of synaptophysin, which carry the risk of being misinterpreted as a MANEC or a NEC. We investigated INSM1 according to the immunoreactive score in our main cohort of 1,033 conventional CRCs and 21 MANECs/NECs in comparison to the expression of synaptophysin and chromogranin A and correlated the results with clinicopathological parameters and patient survival. All MANECs/NECs expressed INSM1, usually showing high or moderate expression (57% high, 34% moderate, and 9% low), which distinguished them from conventional CRCs, which were usually INSM1 negative or low, even if they diffusely expressed synaptophysin. High expression of INSM1 was not observed in conventional CRCs. Chromogranin A was negative/low in most conventional CRCs (99%), but also in most MANECs/NECs (66%). Comparable results were observed in our independent validation cohorts of conventional CRC (n = 274) and MANEC/NEC (n = 19). Similar to synaptophysin, INSM1 expression had no prognostic relevance in conventional CRCs, while true MANEC/NEC showed a highly impaired survival in univariate and multivariate analyses (e.g. disease‐specific survival: p

    الوصف: Abstract Multiple sclerosis (MS) has traditionally been viewed as a chronic inflammatory disease affecting the white matter of the central nervous system. However, over the past two decades, increasing evidence has highlighted the role of gray matter pathology in MS-related disability. Numerous studies have linked the presence of leptomeningeal inflammation to a more severe disease course, underscoring its potential importance as a driver of gray matter pathology in MS. The major components of leptomeningeal inflammation include T cells, B cells, macrophages, follicular dendritic cells, and plasma cells. Since BAFF [B cell-activating factor of the tumor necrosis factor (TNF) family] promotes B cell survival and maturation and is a co-stimulator of T cells, we used anti-BAFF antibody 10F4 as a BAFF antagonist to study its effect on meningeal inflammation and adjacent brain regions in a relapsing–remitting PLP-EAE (rr-EAE) model of multiple sclerosis in SJL/J mice. rr-EAE mice were treated either with anti-BAFF antibody 10F4 or with IgG control antibody. We performed ultra-high field (11.7 T) MRI to identify areas of meningeal inflammation and track them over time in both treatment groups. We also performed histopathological analysis in brain sections of these mice to study the effects of the BAFF antagonist on leptomeningeal inflammation, and hippocampal and cortical neurons and synapses. We observed that BAFF antagonist treatment reduced B cells, T cells, and myeloid cells in regions of meningeal inflammation. Additionally, we noted that BAFF treatment protected against EAE-induced synaptic and neuronal loss in the adjacent cortex and in the CA1, CA3, and dentate gyrus regions of the hippocampus likely due to its effects on meningeal inflammation.

    الوصف: Objetivo: analisar a qualidade de vida de pessoas com úlceras do pé diabético em tratamento ambulatorial. Método: estudo descritivo e transversal, realizado entre fevereiro e abril de 2019, com 50 pessoas com Diabetes Mellitus, acompanhados em ambulatório especializado. Foram aplicados o questionário sociodemográfico e clínico, o instrumento Freiburg Life Quality Assessment Wound Versão Feridas (FLQA-WK), estatística descritiva e os testes de Comparação t de Student e Anova. Resultados: observou-se menor escore no domínio sintomas físicos (1.84) e maior no domínio vida diária (3.52). O escore total de qualidade de vida foi de 2.61, numa escala que varia de um a cinco, com correlação significativa entre as variáveis clínicas, como tempo de diagnóstico de diabetes >10 anos (p-valor=0,005), internações hospitalares (p valor=0,019) e nefropatia (p-valor=0,001). Conclusão: a qualidade de vida foi considerada regular, com alteração no domínio vida diária, sendo influenciada negativamente por variáveis clínicas.

    الوصف: Abstract Background Mechanisms of development and progression of high-grade serous ovarian cancer (HGSOC) are poorly understood. EVI1 and PARP1, part of TGF-ß pathway, are upregulated in cancers with DNA repair deficiencies with DNA repair deficiencies and may influce disease progression and survival. Therefore we questioned the prognostic significance of protein expression of EVI1 alone and in combination with PARP1 and analyzed them in a cohort of patients with HGSOC. Methods For 562 HGSOC patients, we evaluated EVI1 and PARP1 expression by immunohistochemical staining on tissue microarrays with QuPath digital semi-automatic positive cell detection.     الوصف: Due to the growing interest in the use of cannabinoids in supportive therapies, they are increasingly used together with anti-inflammatory drugs. Cannabinoids inhibit gastrointestinal motility, while steroidal and nonsteroidal anti-inflammatory drugs influence motility in other ways. The aim of the research was to study the interactions between cannabidiol (CBD) and these two classes of anti-inflammatory drugs in the context of gastrointestinal motility. Dexamethasone (DEX) was selected as a steroidal drug and diclofenac (DCF) as a nonsteroidal counterpart.
