    الوصف: Melanoma brain metastasis (MBM) frequently occurs in patients with advanced melanoma; yet, our understanding of the underlying salient biology is rudimentary. Here, we performed single-cell/nucleus RNA-seq in 22 treatment-naive MBMs and 10 extracranial melanoma metastases (ECMs) and matched spatial single-cell transcriptomics and T cell receptor (TCR)-seq. Cancer cells from MBM were more chromosomally unstable, adopted a neuronal-like cell state, and enriched for spatially variably expressed metabolic pathways. Key observations were validated in independent patient cohorts, patient-derived MBM/ECM xenograft models, RNA/ATAC-seq, proteomics, and multiplexed imaging. Integrated spatial analyses revealed distinct geography of putative cancer immune evasion and evidence for more abundant intra-tumoral B to plasma cell differentiation in lymphoid aggregates in MBM. MBM harbored larger fractions of monocyte-derived macrophages and dysfunctional TOX+CD8+ T cells with distinct expression of immune checkpoints. This work provides comprehensive insights into MBM biology and serves as a foundational resource for further discovery and therapeutic exploration.

    الوصف: Five years ago, the Melanoma Research Foundation (MRF) conducted an assessment of the challenges and opportunities facing the melanoma research community and patients with melanoma. Since then, remarkable progress has been made on both the basic and clinical research fronts. However, the incidence, recurrence, and death rates for melanoma remain unacceptably high and significant challenges remain. Hence, the MRF Scientific Advisory Council and Breakthrough Consortium, a group that includes clinicians and scientists, reconvened to facilitate intensive discussions on thematic areas essential to melanoma researchers and patients alike, prevention, detection, diagnosis, metastatic dormancy and progression, response and resistance to targeted and immune-based therapy, and the clinical consequences of COVID-19 for patients with melanoma and providers. These extensive discussions helped to crystalize our understanding of the challenges and opportunities facing the broader melanoma community today. In this report, we discuss the progress made since the last MRF assessment, comment on what remains to be overcome, and offer recommendations for the best path forward.

    الوصف: Precision agriculture is a powerful solution to mitigate the environmental impact of agricultural systems, optimize crop inputs and cost reduction. The aim of this work is to analyze the benefits of precision farming practices (irrigation, fertilization and phytosanitary treatment) through a life cycle sustainability assessment (environmental, economic and social impacts) using a detailed life cycle inventory for stone fruit production compared to traditional production. The 8-year life cycle inventory was provided by a local producer in southern Spain. The system boundaries include a "gate to farm gate" approach using 1 kg of stone fruit as the functional unit. The streamlined analysis incorporates environmental, life cycle cost and social risk analysis for this crop production. The results show that the reduced input requirements of the precision agriculture scenario led to lower environmental damage, reduced economic costs and lower social risks, with average impact reductions ranging between 20-30% in most sustainability categories.

    الوصف: For more than three decades, efforts have been made to achieve the consensus that higher education should train professionals with subject-specific knowledge and skills and generic competencies that will enable them to perform successfully in their studies, jobs, and social life contexts. This requires the adoption of curriculum design approaches capable of contextualizing the form and scope of these crucial generic competencies to support both student learning on the course and graduate attributes for employability and career progression. Despite the long history of the concept of generic competencies, the integration of these generic competencies into discipline-specific learning outcomes remains a challenge today. Moreover, it has recently become clear that the definition and approach to generic competencies in higher education requires a context-dependent perspective, given the peculiarities of each subject area. Aware of this difficulty, the Spanish Institute of Engineering has promoted the creation of a forum in which different Spanish higher education institutions involved in engineering participate. The aim of this forum is analysing and improving the treatment of generic skills in these disciplines. The School of Agricultural, Food and Biosystems Engineering (ETSIAAB), one of the institutions participating in the forum, is taking advantage of the analysis of its curricula to initiate a project focused on improving the generic skills defined for their programmes of study.

    الوصف: Electronic feeding stations are frequently used to automatically record feeding behavior in pigs by collecting feed ingested and live mass in each visit. In breeding programmes, the identification of the most efficient animals is a priority objective. To achieve this, they combine the information provided by the electronic feeding stations with that derived from growth models previously adjusted according to the line of animals. It has been found that this practice eliminates infrequent but possible events by substituting intake and body weight values collected by the stations with values interpolated according to the growth models. This results in a double estimation of efficiency, one derived solely from the debugged records of the feeding stations and the other from the growth models. It has been shown that selection of animals according to efficiencies determined only by the growth model leaves out individuals with real efficiencies according to stations above 0.47. Consideration of both efficiencies in the classification of animals would lead to more robust selection procedures.

    الوصف: The combination of visual assessment of whole body [18F]FDG PET images and evaluation of bone marrow samples by Multiparameter Flow Cytometry (MFC) or Next-Generation Sequencing (NGS) is currently the most common clinical practice for the detection of Measurable Residual Disease (MRD) in Multiple Myeloma (MM) patients. In this study, radiomic features extracted from the bone marrow biopsy locations are analyzed and compared to those extracted from the whole bone marrow in order to study the representativeness of these biopsy locations in the image-based MRD assessment. Whole body [18F]FDG PET of 39 patients with newly diagnosed MM were included in the database, and visually evaluated by experts in nuclear medicine. A methodology for the segmentation of biopsy sites from PET images, including sternum and posterior iliac crest, and their subsequent quantification is proposed. First, starting from the bone marrow segmentation, a segmentation of the biopsy sites is performed. Then, segmentations are quantified extracting SUV metrics and radiomic features from the [18F]FDG PET images and are evaluated by Mann–Whitney U-tests as valuable features differentiating PET+/PET− and MFC+ /MFC− groups. Moreover, correlation between whole bone marrow and biopsy sites is studied by Spearman ρ rank. Classification performance of the radiomics features is evaluated applying seven machine learning algorithms. Statistical analyses reveal that some images features are significant in PET+/PET− differentiation, such as SUVmax, Gray Level Non-Uniformity or Entropy, especially with a balanced database where 16 of the features show a p value < 0.001. Correlation analyses between whole bone marrow and biopsy sites results in significant and acceptable coefficients, with 11 of the variables reaching a correlation coefficient greater than 0.7, with a maximum of 0.853.     الوصف: To supply the off-season melon market, Europe imports from distant markets in other countries, mainly Brazil. Cold transportation takes at least 15–20 days, thus increasing the risk of quality losses. Moreover, product deliveries, especially in international markets, can result in supply chain inefficiencies that negatively affect carbon footprint and expected freshness. Implementing quality sensors and advanced cold chain management could help to reduce these problems. The objective of this work was to monitor a real transoceanic intermodal transport of melons (Brazil to Spain), through the implementation of multi-distributed environmental sensors (15 ibuttons loggers) to evaluate the remaining shelf-life (RSHL) of melons at destination. The sensors’ location within the cargo reached a maximum variability range of 4 ◦C. Using digital sensors to track temperature variations, it was verified that in different locations in the container, the melon RSHL at the end of the journey, was nine days and 19 h in colder spots, while in the hottest spot, the RSHL was reduced to five days and 22 h. This fact has substantial implications for improved tracking of temperature to maintain fruit quality for market, potentially reducing waste, and contributing to higher profit margins for international food supply chains.

    الوصف: We are conducting a multicenter study to identify classifiers predictive of disease-specific survival in patients with primary melanomas. Here we delineate the unique aspects, challenges, and best practices for optimizing a study of generally small-sized pigmented tumor samples including primary melanomas of at least 1.05mm from AJTCC TNM stage IIA-IIID patients. We also evaluated tissue-derived predictors of extracted nucleic acids’ quality and success in downstream testing.
