الوصف: Among different staining methods used to demonstrate the nuclear abnormalities, Feulgen is one of the most reliable method. Feulgen staining is specific, sensitive method for evaluating the DNA damages.It has been shown that using non-DNA specific stains for monitoring the nuclear anomalies lead to false-positive or false-negative results. From self-experience, immersing the stained slides in hematoxylin for 3-5 minutes in final step after staining the slides with light green, provides a greater ground contrast and very better visualizing of nucleus. By this method, a clear distinction can be made between micronuclei and other structures suggestive of a micronucleus.

الوصف: The genus Polianthes belongs to the subfamily Agavoideae of the Asparagaceae family formerly known as Agavaceae. The genus is endemic to México and comprises about 15 species, among them is Polianthes tuberosa L. The aim of this work was to study and characterize the embryo sac and early embryo development of this species in order to generate basic knowledge for its use in taxonomy, in vitro fertilization and production of haploid plants and to complement studies already performed in other genera and species belonging to the Agavoideae sub-family. It was found that the normal development of the P. tuberosa var. Simple embryo sac follows a monosporic pattern of the Polygonum type and starts its development from the chalazal megaspore. At maturity, the embryo sac is of a pyriform shape with a chalazal haustorial tube where the antipodals are located, just below the hypostase, which connects the embryo sac with the nucellar tissue of the ovule. The central cell nucleus shows a high polarity, being located at the chalazal extreme of the embryo sac. The position of cells inside the P. tuberosa embryo sac may be useful for in depth studies about the double fertilization. Furthermore, it was possible to make a chronological description of the events that happen from fertilization and early embryo development to the initial development of the endosperm which was classified as of the helobial type.

الوصف: Although many papers dealing with the description of new ciliate taxa are published each year, species taxonomy and identification in most groups of the phylum Ciliophora remain confused. This is largely due to a scarcity of surveys on the systematics of immediately higher levels (genera and families) providing data for old and new species together. Spirostomum is a common and distinctive inhabitant of fresh- and brackish water environments, including artificial and eutrophic ones, and is a good model for applied ecology and symbiosis research. Despite this, only 3 of the numerous species are commonly cited, and no studies have yet confirmed their monophyly, with the consequence that reproducibility of the results may be flawed. In this paper we present morphological and molecular data for 30 Spirostomum populations representing 6 different morphospecies, some of which were collected in previously unreported countries. We performed a detailed revision of Spirostomum systematics combining literature surveys, new data on hundreds of organisms and statistical and phylogenetic analyses; our results provide insights on the evolution, ecology and distribution of known morphospecies and a novel one: Spirostomum subtilis sp. n. We also offer tools for quick species identification. © 2014 Elsevier GmbH.

الوصف: Background Early-stage oral squamous cell carcinoma (OSCC) patients have a one-in-four risk of regional metastasis (LN+), which is also the most significant prognostic factor for survival. As there are no validated biomarkers for predicting LN+ in early-stage OSCC, elective neck dissection often leads to over-treatment and under-treatment. We present a machine-learning-based model using the quantitative nuclear phenotype of cancer cells from the primary tumor to predict the risk of nodal disease. Methods and findings Tumor specimens were obtained from 35 patients diagnosed with primary OSCC and received surgery with curative intent. Of the 35 patients, 29 had well (G1) or moderately (G2) differentiated tumors, and six had poorly differentiated tumors. From each, two consecutive sections were stained for hematoxylin & eosin and Feulgen-thionin staining. The slides were scanned, and images were processed to curate nuclear morphometric features for each nucleus, measuring nuclear morphology, DNA amount, and chromatin texture/organization. The nuclei (n = 384,041) from 15 G1 and 14 G2 tumors were randomly split into 80% training and 20% test set to build the predictive model by using Random Forest (RF) analysis which give each tumor cell a score, NRS. The area under ROC curve (AUC) was 99.6% and 90.7% for the training and test sets, respectively. At the cutoff score of 0.5 as the median NRS of each region of interest (n = 481), the AUC was 95.1%. We then developed a patient-level model based on the percentage of cells with an NRS ≥ 0.5. The prediction performance showed AUC of 97.7% among the 80% (n = 23 patient) training set and with the cutoff of 61% positive cells achieved 100% sensitivity and 91.7% specificity. When applying the 61% cutoff to the 20% test set patients, the model achieved 100% accuracy. Conclusions Our findings may have a clinical impact with an easy, accurate, and objective biomarker from routine pathology tissue, providing an unprecedented opportunity to improve neck management decisions in early-stage OSCC patients.

الوصف: Haemic neoplasia (HN) in bivalves has been reported in association with mass mortality events in various species of molluscs. The aim of this work was to quantify the nuclear morphometry and DNA content of neoplastic cells of mussels Mytilus galloprovincialis affected by HN using nuclear densitometry in Feulgen-stained preparations. The results were also compared with a population of normal mussel haemocytes. We captured 256 images of 3 different neoplasia stages and 120 images of normal haemocytes; thus, a total of 120,166 nuclei were analysed. الوصف: Context: Abnormalities in nuclear morphology are very frequently seen in dysplasia, such as nuclear area, diameter, shape, number of nucleoli and membrane outline. The purpose of this study was to observe and compare the nuclear features in different grades of epithelial dysplasia in leukoplakia and to evaluate the use of Feulgen stain for observing the nuclear features in oral epithelial dysplasia in leukoplakia. Seventy paraffin embedded tissue section (20 mild, 20 moderate, 20 severe dysplasia cases and 10 control specimens) were analyzed for nuclear morphology using Feulgen stain under trinocular research microscope. Statistically significant results were obtained with P > 0.001, when intergroup comparison was done except in case of nuclear area and diameter between mild and moderate dysplasia. Nuclear features reflect cell's biological potential and its morphometry was found to be a useful tool for differentiating different grades of dysplasia.
