    الوصف: The increasing importance of fungal infections has fueled the search for new beneficial alternatives substance from plant extracts. The current study investigates the antifungal and antibiofilm activity of Inula viscosa (L.) Aiton and Globularia alypum (L.) leaves extracts against Candida both in vitro and in vivo. The inhibition of planktonic and sessile Candida albicans and Candida glabrata growth using both leaf extracts are evaluated. Moreover; an in vivo infection model using Galleria mellonella larvae; infected and treated with the extracts are performed. All extracts show fungicidal activity; with a minimum fungicidal concentration (MFC) ranging from 128 to 512 mu g mL(-1) against the two selected strains of Candida. In particular, the best results are obtained with methanolic extract ofI. viscosa and G. alypum with an MFC value of 128 mu g mL(-1). The extracts are capable to prevent 90% of biofilm development at minor concentrations ranging from 100.71 +/- 2.49 mu g mL(-1) to 380.4 +/- 0.92 mu g mL(-1). In vivo, tests on Galleria mellonella larvae show that the extracts increase the survival of the larvae infected with Candida. The attained results reveal that I. viscosa and G. alypum extracts may be considered as new antifungal agents and biofilm inhibiting agents for the pharmaceutical and agro-food field.

    الوصف: This work aimed to investigate Sinapis pubescens subsp. pubescens spontaneously grown in Sicily (Italy) as new potential source of active metabolites; specifically, a comparative study on leaf, flower, and stem hydroalcoholic extracts was performed. Polyphenols were quantitatively determined by spectrophotometric methods and characterized by HPLC-PDA/ESI-MS; a total of 55 polyphenolic compounds were identified, highlighting considerably different qualitative-quantitative profiles. The extracts showed antioxidant activity, evaluated by in vitro assays; particularly, the leaf extract displayed the best radical scavenging activity (DPPH test) and reducing power, while the flower extract showed the greatest chelating activity. The antimicrobial properties of the extracts were investigated against bacteria and yeasts by standard methods; no antimicrobial activity was found against the strains tested. The extracts resulted to be non-toxic after preliminary toxicity evaluation by the Artemia salina lethality bioassay. The aerial parts of S. pubescens subsp. pubescens proved to be valuable sources of antioxidants for pharmaceutical and nutraceutical applications.

    الوصف: The potential of bergamot pomace for the development of materials able to efficiently remove cadmium(II) from water and for the recovery of bioactive compounds has been explored. To this purpose, raw bergamot waste resulting after industrial essential oil and juice extraction was mechanically ground, desiccated, pretreated with various chemicals (e.g. NaOH, HNO3, H2O2, H2O, 2-propanol) and dried up to constant weight thus affording solid samples that were characterized by ATR FT-IR spectroscopy. The solutions recovered after the pomace pretreatments were investigated by means of HPLC in combination with PDA and MS detectors to assess the residual content of bioactive components, e.g., phenolic and oxygenated heterocyclic compounds (OHCs). Potentiometric studies were performed on suspensions at t = 25 °C, I = 0.10 mol dm−3 in NaNO3(aq) to investigate pomace acid-base properties and binding ability towards Cd2+ ions. Sorption efficiency was investigated by means of kinetic and isotherm batch experiments and resulted to be 92 ± 7 mg g−1. Once loaded, sorbent reusability was tested by performing metal stripping cycles using various desorbents (HCl, HNO3, L-GLDA, S,S-EDDS, EDTA) with an efficiency of ∼ 60% after one cycle. The equilibrium Cd2+ concentration in solution was determined by differential pulse voltammetry and ICP-OES.

    الوصف: In this study, the chemical compositions of essential oils obtained from three plants, namely Salvia officinalis, Lavandula angustifolia, and Mentha pulegium, along with their insecticidal activities and feeding deterrence against Plodia interpunctella, were investigated. The chemical composition of S. officinalis essential oil was majorly dominated by α-thujone (26.99%) and camphor (25.50%); on the other hand, limonene (66.12%), eucalyptol (66.12%), and camphor (14.31%) were the major compounds of L. angustifolia essential oil whereas for M. pulegium EO a dominance of pulegone (83.06%) was attained. M. pulegium essential oil showed important contact toxicity (100%) at 3 hours of exposure with the 0.25 μL/cm2 concentration, followed by S. officinalis and L. angustifolia with 90% of mortality reported with the highest concentration of 0.50% within 3 hours. A relative growth rate (RGR) of 0.08 mg/mg/day and -0.31 mg/mg/day was recorded with S. officinalis and M. pulegium oils, respectively, with the highest concentration of 32 μL/pellet. With L. angustifolia the lowest RGR of -0.08 mg/mg/day was attained with the 16 μl/pellet concentration. The food deterrence index experienced an increase to 89% with S. officinalis and L. angustifolia oils for the three concentrations; with M. pulegium, 68.18% was recorded with the 32 μL/pellet. In conclusion, this study suggests that these essential oils could potentially serve as an effective contact toxicant and a potent eco-friendly inhibitory of the feeding behavior of Plodia interpunctella larvae.

    الوصف: Background: Inflammation is a natural response of the body to tissue injury or infection, but chronic inflammation can contribute to the development of various diseases. Therefore, finding effective ways to reduce inflammation is of great interest to researchers and healthcare providers. The aim of the study was the elucidation of the phenolic profile from J. oxycedrus leaves and berries followed by in vitro antioxidant and anti-inflammatory assays. Despite the existence of some works on this species, most of these researchers focus their attention on cade oil and only few were interested to study leaves and berries extracts. Methods: HPLC-DAD-ESI/MS2 was used for their characterization of the phenolic profile whereas spectrophotometric microassays in non-cellular and cellular systems (RAW 264.7 macrophages cell line) was assessed for their in vitro antioxidant and anti-inflammatory potentials. Results: Up to twenty-one polyphenolic compounds were positively identified in both extracts. Both leaves and berries extracts showed strong antioxidant capacity to scavenge O2●, with IC50 values of 22.86 ± 2.40 and 122.62 ± 9.42 μg/mL. Moreover, both revealed to have potential to inhibit 5-lipoxygenase in a dose-dependent way, being leaves the most effective vegetal material. Conclusion: The results suggest that J. oxycedrus leaves and berries could be considered exquisite sources of bioactive molecules for a potential further application in the food and pharmaceutical industries.

    الوصف: A rapid and practicable analytical method for the measurement of short-chain fatty acids (SCFAs) in human plasma was developed. The extraction procedure involved the use of acidified water and methyl tert-butyl ether (MTBE), while the separation and detection of SCFAs, including acetic, propionic, and butyric acids was carried out by using gas chromatography-mass spectrometry (GC–MS) technique. The novelty of the research involves reducing the analysis time (less than 7 min) by using the novel fast GC–MS method. A narrow-bore GC capillary column of dimensions 30 m × 0.25 mm ID × 0.25 μm df with acid-modified poly(ethylene glycol) stationary phase was employed for the chromatographic separation. The signals of target compounds were acquired in selected ion monitoring (SIM) mode monitoring a quantifier ion (Q) and two qualifier ions (q1 and q2). Linearity of the method, limits of detection (LoD) and quantification (LoQ) were evaluated. In detail, regression coefficients of the calibration curves were between 0.9960 and 0.9933; LoDs ranged from 0.02 μM to 0.03 μM, while LoQs from 0.06 μM to 0.10 μM.

    الوصف: Acinos alpinus and Ziziphora hispanica belonging to the Lamiaceae family are well-known for their biological properties. The phenolic composition was characterized using HPLC coupled to photodiode array and electrospray ionization mass spectrometry. The antioxidant capacity was assessed by 2,2′-diphenyl-1-picrylhydrazyl radical and ferric reducing antioxidant power assays. The antibacterial activity was evaluated by disc-diffusion and broth micro-dilution methods. A total of 53 phenolic compounds were identified in both extracts, with the predominance of caffeoylquinic acid isomers with a content of 2.09 mg/g in A. alpinus extract, while coumarylquinic acid isomers with a content of 17.33 mg/g was determined in Z. hispanica extract. The Acinos alpinus extract displayed the highest radical scavenging activity (IC50 DPPH = 3.226±0.035 mg/mL) and reducing power (EC50 FRAP = 3.792±0.001 mg/mL), followed by Z. hispanica extract with IC50 DPPH value of 7.265±0.209 mg/mL, and EC50 FRAP value of 24.689± 0.106 mg/mL. Furthermore, both A. alpinus and Z. hispanica extracts exhibited a bactericidal effect against six pathogenic bacteria with a MIC value from 4.16 to 33.33 mg/mL. On the basis of the results achieved, the aerial parts of Moroccan Acinos alpinus and Ziziphora hispanica might be used as natural agents in the food and pharmaceutical field.

    الوصف: There is substantial experimental evidence for dysregulation of several microRNA (miRNA) expression levels in Alzheimer’s disease (AD). MiRNAs modulate critical brain intracellular signaling pathways and are associated with AD core pathophysiological mechanisms. First, we conducted a real-time quantitative PCR-based pilot study to identify a set of brain-enriched miRNAs in a monocentric cohort of cognitively normal individuals with subjective memory complaints, a condition associated with increased risk of AD. Second, we investigated the impact of age, sex, and the Apolipoprotein E ε4 (APOE ε4) allele, on the identified miRNA plasma concentrations. In addition, we explored the cross-sectional and longitudinal association of the miRNAs plasma concentrations with regional brain metabolic uptake using amyloid-β (Aβ)-positron emission tomography (Aβ-PET) and 18F-fluorodeoxyglucose-PET (18F-FDG-PET). We identified a set of six brain-enriched miRNAs—miRNA-125b, miRNA-146a, miRNA-15b, miRNA-148a, miRNA-26b, and miRNA-100. Age, sex, and APOE ε4 allele were not associated with individual miRNA abundance. MiRNA-15b concentrations were significantly lower in the Aβ-PET-positive compared to Aβ-PET-negative individuals. Furthermore, we found a positive effect of the miRNA-15b*time interaction on regional metabolic 18F-FDG-PET uptake in the left hippocampus. Plasma miRNA-125b concentrations, as well as the miRNA-125b*time interaction (over a 2-year follow-up), were negatively associated with regional Aβ-PET standard uptake value ratio in the right anterior cingulate cortex. At baseline, we found a significantly negative association between plasma miRNA-125b concentrations and 18F-FDG-PET uptake in specific brain regions. In an asymptomatic at-risk population for AD, we show significant associations between plasma concentrations of miRNA-125b and miRNA-15b with core neuroimaging biomarkers of AD pathophysiology.     الوصف: The traditional approach to biomarker discovery for any pathology has been through hypothesis-based research one candidate at a time. The objective of this study was to develop an agnostic approach for the simultaneous screening of plasma for consistent molecular differences between a group of individuals exhibiting a pathology and a group of healthy individuals. To achieve this, we focused on developing a predictive tool based on plasma for the amount of brain amyloid-β deposition as observed in PET scans. The accumulation of brain amyloid-β (Aβ) plaques is a key risk factor for the development of Alzheimer’s disease. A contrast was established between cognitively normal individuals above the age of 70 that differed for the amount of brain amyloid-β observed in PET scans (INSIGHT study group). Positive selection was performed against a pool of plasma from individuals with high brain amyloid and negative selection against a pool of plasma from individuals with low brain amyloid This enriched, selected library was then applied to plasma samples from 11 individuals with high levels of brain amyloid and 11 individuals with low levels of brain Aβ accumulation. Each of these individually selected libraries was then characterized by next generation sequencing, and the relative frequency of 10,000 aptamer sequences that were observed in each selection was screened for ability to explain variation in brain amyloid using sparse partial least squares discriminant analysis. From this analysis a subset of 44 aptamers was defined, and the individual aptamers were synthesized. This subset was applied to plasma samples from 70 cognitively normal individuals all above the age of 70 that differed for brain amyloid deposition. 54 individuals were used as a training set, and 15 as a test set.
