    مستخلص: Detailed understanding of crustal components and tectonic history of forearcs is important due to their geological complexity and high seismic hazard. The principal component of the Cascadia forearc is Siletzia, a composite basaltic terrane of oceanic origin. Much is known about the lithology and age of the province. However, glacial sediments blanketing the Puget Lowland obscure its lateral extent and internal structure, hindering our ability to fully understand its tectonic history and its influence on modern deformation. In this study, we apply map‐view interpretation and two‐dimensional modeling of aeromagnetic and gravity data to the magnetically stratified Siletzia terrane revealing its internal structure and characterizing its eastern boundary. These analyses suggest the contact between Siletzia (Crescent Formation) and the Eocene accretionary prism trends northward under Lake Washington. North of Seattle, this boundary dips east where it crosses the Kingston arch, whereas south of Seattle the contact dips west where it crosses the Seattle uplift (SU). This westward dip is opposite the dip of the Eocene subduction interface, implying obduction of Siletzia upper crust at this southern location. Elongate pairs of high and low magnetic anomalies over the SU suggest imbrication of steeply‐dipping, deeply rooted slices of Crescent Formation within Siletzia. We hypothesize these features result from duplication of Crescent Formation in an accretionary fold‐thrust belt during the Eocene. The active Seattle fault divides this Eocene fold‐thrust belt into two zones with different structural trends and opposite frontal ramp dips, suggesting the Seattle fault may have originated as a tear fault during accretion. Plain Language Summary: The Puget Lowland of Washington State contains several potentially dangerous seismic faults, including the Seattle fault, which runs south of downtown Seattle. To accurately assess the earthquake hazard in this region, we need to understand the architecture and geologic history of the rocks that host these faults, deep below the Puget Lowland. We do this by using small changes in Earth's gravity and magnetic fields to create images of the Earth's subsurface. These rocks formed in a subduction zone 50 million years ago when a set of volcanic islands, similar to modern day Iceland, collided with the edge of North America. This added a layer of rock, called Siletzia, to the continent. We show that as the islands piled up, they broke and folded into mountain ranges. South of Seattle, Siletzia was pushed up and over ancient North America, whereas to the north Siletzia was pulled down and under the continent. We argue that a tear in Siletzia between these two zones eventually became the proto‐Seattle fault, which provides a story for the Seattle fault's origin and earliest history. Our images also provide information that can improve models of ground shaking from future earthquakes affecting the greater Seattle urban area.     مستخلص: Estimation of probable maximum flood (PMF) is a crucial process in water resources management and in the design of large hydraulic structures. However, there are uncertainties in the estimation of hydrologic conditions that contribute to extreme floods. In particular, this is the case in snow‐dominated regions, as surface air temperature and wind speed are understood to have a substantial effect on the magnitude of a flood during a storm event. Motivated by the development of a new approach to investigate and estimate reliable PMF values and in an attempt to resolve the uncertainty issues, this study introduces a physically based modeling approach. For the case study, seven watersheds located in the Sierra‐Nevada mountain range of California, including Cosumnes, Mokelumne, Stanislaus, Tuolumne, Merced, Upper San Joaquin, and Upper Kings were selected. The hydroclimate model was first implemented over the physical boundaries of the study region, and then utilized to simulate possible maximum flood conditions with input from 10 extreme precipitation scenarios. The study results provide evidence of a nonlinear atmospheric–hydrologic system; the extreme 72‐h basin‐averaged precipitation depth was found not to be linearly proportional to 72‐h flow volume equivalent depth. It can also be concluded that a large precipitation depth may not be the sole reason for a large flood event. Temperature and other atmospheric variables also contribute significantly to the production of snowfall and liquid water available for runoff, and to the resulting hydrologic response, such as the flood peak discharge and volume. Participants with no hypertension diagnosis or medications and elevated blood pressure (BP) were randomized to one of three diagnostic regimens: (i) Clinic, (ii) Home, or (iii) Kiosk. Participants completed questionnaires at baseline, after completion of the diagnostic regimens, and at 6 months. Outcomes included changes from baseline in health-related quality of life (HRQOL), BP-related worry, and thoughts about having a stroke or heart attack. RESULTS Participants (n  = 482) were mostly over age 50 (77.0%), and White race (80.3%). HRQOL did not significantly change from baseline to 3 weeks or 6 months. Among all participants, BP-related worry and concerns about having a heart attack or stroke increased significantly from baseline to 3 weeks, with heart attack and stroke concerns significantly higher in the Kiosk compared Clinic and Home groups. At 6 months, thoughts about having a heart attack or stroke returned to baseline overall and in the Kiosk group, however BP-related worry was significantly higher among those with, compared to those without, a new hypertension diagnosis. CONCLUSIONS The hypertension diagnostic process did not lead to short-term or intermediate-term changes in self-reported HRQOL. However, BP-related worry increased short-term and persisted at 6 months among individuals with a new hypertension diagnosis. Results warrant validation in more representative populations and additional exploration of the impacts of this worry on psychological well-being and hypertension control.     الوصف: Hippo signaling has been recognized as a key tumor suppressor pathway. Here, we perform a comprehensive molecular characterization of 19 Hippo core genes in 9,125 tumor samples across 33 cancer types using multidimensional “omic” data from The Cancer Genome Atlas. We identify somatic drivers among Hippo genes and the related microRNA (miRNA) regulators, and using functional genomic approaches, we experimentally characterize YAP and TAZ mutation effects and miR-590 and miR-200a regulation for TAZ. Hippo pathway activity is best characterized by a YAP/TAZ transcriptional target signature of 22 genes, which shows robust prognostic power across cancer types. Our elastic-net integrated modeling further reveals cancer-type-specific pathway regulators and associated cancer drivers. Our results highlight the importance of Hippo signaling in squamous cell cancers, characterized by frequent amplification of YAP/TAZ, high expression heterogeneity, and significant prognostic patterns. This study represents a systems-biology approach to characterizing key cancer signaling pathways in the post-genomic era.     الوصف: We performed an extensive immunogenomic analysis of more than 10,000 tumors comprising 33 diverse cancer types by utilizing data compiled by TCGA. Across cancer types, we identified six immune subtypes—wound healing, IFN-γ dominant, inflammatory, lymphocyte depleted, immunologically quiet, and TGF-β dominant—characterized by differences in macrophage or lymphocyte signatures, Th1:Th2 cell ratio, extent of intratumoral heterogeneity, aneuploidy, extent of neoantigen load, overall cell proliferation, expression of immunomodulatory genes, and prognosis. Specific driver mutations correlated with lower (CTNNB1, NRAS, or IDH1) or higher (BRAF, TP53, or CASP8) leukocyte levels across all cancers. Multiple control modalities of the intracellular and extracellular networks (transcription, microRNAs, copy number, and epigenetic processes) were involved in tumor-immune cell interactions, both across and within immune subtypes. Our immunogenomics pipeline to characterize these heterogeneous tumors and the resulting data are intended to serve as a resource for future targeted studies to further advance the field. Thorsson et al. present immunogenomics analyses of more than 10,000 tumors, identifying six immune subtypes that encompass multiple cancer types and are hypothesized to define immune response patterns impacting prognosis. This work provides a resource for understanding tumor-immune interactions, with implications for identifying ways to advance research on immunotherapy.
