    الوصف: Membrane contact sites (MCS) circumvent the topological constraints of functional coupling between different membrane-bound organelles by providing a means of communication and exchange of materials. One of the most characterised contact sites in the cell is that between the endoplasmic reticulum and the mitochondrial (ERMCS) whose function is to couple cellular Ca 2+ homeostasis and mitochondrial function. Inositol 1,4,5-trisphosphate receptors (IP 3 Rs) on the ER, glucose-regulated protein 75 (GRP 75) and voltage-dependent anion channel 1 (VDAC1) on the outer mitochondrial membrane are the canonical component of the Ca 2+ transfer unit at ERMCS. These are often reported to form a Ca 2+ funnel that fuels the mitochondrial low-affinity Ca 2+ uptake system. We assess the available evidence on the IP 3 R subtype selectivity at the ERMCS and consider if IP 3 Rs have other roles at the ERMCS beyond providing Ca 2+ . Growing evidence suggests that all three IP 3 R subtypes can localise and regulate Ca 2+ signalling at ERMCS. Furthermore, IP 3 Rs may be structurally important for assembly of the ERMCS in addition to their role in providing Ca 2+ at these sites. Evidence that various binding partners regulate the assembly and Ca 2+ transfer at ERMCS populated by IP 3 R-GRP75-VDAC1, suggesting that cells have evolved mechanisms that stabilise these junctions forming a Ca 2+ microdomain that is required to fuel mitochondrial Ca 2+ uptake.

The orientation of the spindle, guided by internal and external cues, determines the axis of cell division and thereby contributes to tissue morphogenesis. Progression through mitosis requires local Ca2+ signals at critical steps, and because store-operated Ca2+ entry is inhibited during mitosis, those signals probably require Ca2+ release through inositol 1,4,5-trisphosphate receptors (IP3Rs). In cells without IP3Rs, astral microtubules around the daughter centrosome are shorter than those at the mother centrosome, and the mitotic spindle fails to align with the substratum during metaphase. The misalignment is due to the spindle ineffectively detecting internal cues rather than a failure of cells to recognize the substratum. Expression of type 3 IP3R is sufficient to rescue spindle alignment, but only if the IP3R has a functional pore. We conclude that Ca2+ signals evoked by IP3Rs are required to orient the mitotic spindle.

    الوصف: The article is addressed to a wide range of specialists involved in data science including ontological data modeling and knowledge extracting in the various fields related to sustainable development (SD). A hierarchically structured ontological metamodel of SD is developed consisting of a conceptual layer of interconnected common, domain and application ontologies, and a physical layer of instance databases and documents. The SD common ontology is the top level of the conceptual layer and is associated with the next level of the SD domain ontologies in the economy, society, and nature. The third level is of the SD application ontologies and is illustrated by an example application ontology of firm sustainability. All ontologies of the SD metamodel have links to the relevant external ontologies and to specific instance databases. The proposed metamodel is a useful tool for SD knowledge structuring and classification, as well as a base for building machine learning models as a part of artificial intelligence.

    الوصف: Sustainable development in its various aspects is a multifaceted and contradictory concept, interpreted by multiple sources. This study exposes the need to explore and develop a productive coevolutionary approach to build modular ontological metamodel of sustainable development including a set of interconnected ontologies and instance databases. Based on a review and on an analysis of existing ontologies in this area, a methodology for building common ontology of sustainable development is proposed as a top level of the metamodel. Using terminology from well-established and authoritative sources in the area, as well as the methods, the resources, and the standards of Semantic Web, the methodology contains stages: exploring the SD concepts; controlled vocabulary and thesaurus creating; ontology formalization, verification, querying, and publication. Relationships with the respective domain and other related to sustainable development ontologies are established. In conclusion, the results and the future development of the ontological metamodel are discussed.

    الوصف: Variability in the glycosylation profile of therapeutic monoclonal antibodies (mAbs), due to recombinant production technologies, leads to inconsistencies in effector functions and pharmacokinetic properties, both batch-to-batch and within single batches. It also poses regulatory concerns over the effectiveness of commercially available formulations. In vitro chemoenzymatic glycoengineering of variants displaying a homogeneous glycan profile is a trending strategy for ensuring consistent, controlled, and enhanced therapeutic performance, but reported successes are largely limited to small-scale applications. The major challenges for the industrial-scale introduction of the technique stem from the need for activated sugar donors, which can participate in undesired side reactions, and from the economic cost of the additional enzymatic steps and purification stages. While recent developments within the area address some of these obstacles, it appears that more effort is required in order to access the untapped potential of biocatalysis to enable the robust production of therapeutically superior constructs.

    الوصف: The social sustainability is one of the three pillars of the sustainable development. In the article the respective domain ontology is developed as a part of the existing ontological metamodel of the sustainable development. A methodology for the ontology building is proposed including concept exploration, controlled vocabulary and thesaurus creating; ontology formalization, verification, querying and publication. The main concepts of the ontology are extracted from a text corpus covering well-established and authoritative sources, approved articles, standards and commonly accepted regulations of United Nations. For the purpose, manual selection and automatic text mining by the language processing toolkit GATE is performed. The ontology is formalized by OWL in Protege environment and after verification its code in GitHub is published.

    الوصف: In the article the problems related to the analysis and measurement of competitiveness at the firm level are explored. An original methodology is proposed on the base of detailed literature review in the area including investigation of the known theoretical studies and the practical achievements. A special attention to the definitions and terminology is paid. The methodology is on the base of existing methods and own algorithms. It proposes an innovative information model of firm competitiveness using ontology engineering for creating metamodel, statistical methods for identifying firm competitiveness criteria and indicators and web technologies for prototype online information system building. A regression analysis is applied revealing the mutual dependency between the firm and the product competitiveness for identifying firm competitiveness criteria and indicators. Based on the methodology a prototype online information system is developed intended for firm competitiveness index calculation and analysis. The proposed information model and the prototype online system fill a gap in the firm competitiveness research and analysis tools. It is expected that their application in the practice would improve awareness of firm managers about positions of their organizations and increase the conclusion quality of the business analysts.

    Lagos-Cabré et al. show that Ca2+ release by IP3 receptors (IP3Rs) regulates spindle orientation during metaphase. Spindles misalign in cells without IP3Rs. Alignment is restored by expression of IP3Rs but not by IP3Rs that cannot release Ca2+. Ca2+ release by IP3Rs regulates astral microtubules and thereby spindle alignment.
