مستخلص: In drinking water science, as in life, there are typically several doorways one can go through to reach an objective. A case in point is the Information Collection Rule (ICR) method for determining concentrations of Cryptosporidium oocysts and Giardia cysts in drinking water. When this method proved to be technically difficult and problematic-with low and highly variable recoveries, frequent false-positive andfalse-negative results, and poor precision and accuracy-even for expert laboratories, the US Environmental Protection Agency (USEPA) closed the door on it and opened another to 'Method 1622: Cryptosporidiumin Water by Filtration/IMS/FA.' This article discusses how method 1622 was developed, tested, and validated for use in USEPA's supplemental surveys to gather data on Cryptosporidium occurrence in US source water. The two laboratories charged with developing, optimizing, and testing method 1622 did so by systematically creating and then evaluating each step in the method for its ability to recover spiked oocysts in reagent water. The laboratories' strategy included selecting an optimal sample size, determining spike dosages, selecting appropriatescientific equipment, conducting initial precision and recovery studies, and determining method detection limits. When they were finished, method 1622 was ready to undergo a validation study. The method, which was subsequently validated, has proved to be a significant improvement over the ICR method. It is faster and easier to perform, and recovery rates are high and consistent-even among laboratories. USEPA also altered method 1622 to create 'Method 1623: Giardia and Cryptosporidium in Water by Filtration/IMS/FA.' This new, fully validated method promises to enable simultaneous recovery and detection of crypto and Giardia, thus opening yet another door to safer drinking water. [ABSTRACT FROM AUTHOR]
