الوصف: Simple Summary Intramuscular fat or fatty acids content was regarded as a quality index of meat as it increases meat tenderness and flavor. In Korea, commercial Hanwoo farms have been supplying concentrate feeds that are 2–3% higher in crude protein (CP) content rather than conventional feeds throughout the entire program and have tried to shorten the feeding period to less than 28 months of age. This has led to a probable stability in lean-to-fat ratio in Hanwoo steers from 19 to 21 months of age. This stability could be linked to the regulation of the gene expression mediating lipogenesis during this phase. However, there is a lack of data regarding the effects of the protein level in isoenergetic concentrate diets on transcriptional activity of such genes at this phase. The conventional feeding program and the current one with relatively higher CP content were compared in two groups of twenty Hanwoo steers. Results showed that higher CP endorsement during the growing phase increased the expression of intramuscular PPARα (p < 0.1) and LPL (p < 0.05) and decreased the genes, such as VLCAD (p < 0.01), GPAT1 (p = 0.001), and DGAT2 (p = 0.016), which are involved in lipogenesis and fatty acid esterification. This may result in a relatively lower lipid turnover which could be responsible for shortening the feeding period. Abstract The present study evaluated the influence of dietary protein level on growth performance, fatty acid composition, and the expression of lipid metabolic genes in intramuscular adipose tissues from 18- to 23-month-old Hanwoo steers, representing the switching point of the lean-to-fat ratio. Forty steers with an initial live weight of 486 ± 37 kg were assigned to one of two treatment groups fed either a concentrate diet with 14.5% CP and or with 17% CP for 6 months. Biopsy samples of intramuscular tissue were collected to analyze the fatty acid composition and gene expression at 23 months of age. Throughout the entire experimental period, all steers were restrained twice daily to allow individual feeding. Growth performance, blood metabolites, and carcass traits, according to ultrasonic measurements, were not affected by the experimental diets. The high-protein diet significantly increased the expression of intramuscular PPARα (p < 0.1) and LPL (p < 0.05) but did not affect genes involved in fatty acid uptake (CD36 and FABP4) nor lipogenesis (ACACA, FASN, and SCD). In addition, it downregulated intramuscular VLCAD (p < 0.01) related to lipogenesis but also GPAT1 (p = 0.001), DGAT2 (p = 0.016), and SNAP23 (p = 0.057), which are involved in fatty acid esterification and adipocyte size. Hanwoo steers fed a high-protein diet at 18–23 months of age resulted in a relatively lower lipid turnover rate than steers fed a low-protein diet, which could be responsible for shortening the feeding period.

الوصف: Objective: This study was conducted to evaluate changes in nutritional value and in situ dry matter (DM) degradability of oak and pine wood before and after steam-digestion process (60 min/160°C/6 atm) and feeding effect of the oak roughage on performance and behavior of Hanwoo steers.Methods: Chemical composition and tannin concentration were analyzed for oak and pine trees before and after the pretreatment. In situ DM and effective degradability of these samples were assessed using a nylon bag method. In vivo trial was performed to estimate animal performance and behavior, using steers fed total mixed ration (TMR) diets containing 0% (control), 25% (OR-25), and 50% (OR-50) of the oak roughage. Eighteen steers were allocated into nine pens (2 steers/pen, 3 pens/treatment) for 52 days according to body weight (BW) and age.Results: By the steam-digestion treatment, the neutral detergent-insoluble fiber was decreased from 86.5% to 71.5% for oak and from 92.4% to 80.5% for pine, thereby increasing non-fiber carbohydrate. In situ DM degradability of treated oak reached 38% at 72 h, whereas that of untreated oak was only 11.9%. The 0 h degradability of the treated pine increased from 5.9% to 12.1%, but the degradability was unchanged thereafter. Animal performance including BW, average daily gain, DM intake, and feed conversion ratio was not different among control and oak treatments. No differences were detected in animal behavior such as lying, standing, rumination, drinking, and eating, except walking. Walking was higher in control than oak treatments with numerically higher eating and lower lying times, probably due to bulkier characteristics of rice straw in the diet.Conclusion: This study demonstrates that the oak roughage can be substituted for 50% of total forage or 100% of rice straw in TMR diets at early fattening stage of Hanwoo steers.

الوصف: An experiment was conducted to evaluate the effects of supplementing feed additives of Barodon®, effective microorganism® (Bacillus (B.) subtilis), and Ampbio® on the growth performance, blood metabolites, stress, and reproductive hormone levels of Korean native heifers. A total of 48 Korean native heifers were assigned to four groups with 6 heifers in each group. The groups were control, Barodon (0.2%), beneficial microorganism (1%), and Ampbio (1%). Animals in all groups were fed a basal diet composed of selected feed additives and water ad libitum for 99 days. Results showed that there are significant changes in body weight and daily gain in the Ampbio-supplemented group as compared with the control and other feed additives groups (p 0.05). The increased level of feed intake (7.30 ± 0.03 kg) and feed requirements (10.81 ± 0.52 kg) was observed in the Ampbio-fed group followed by the effective microorganism (EM), Barodon, and control groups. There were no significant changes in albumin, glucose, glutamic oxaloacetic transaminase (SGOT), serum pyruvic transaminase (SGPT), and total protein level, but the decreased levels of total cholesterol and triglycerides and the increased level of blood urea nitrogen were noted in the Ampbio-fed group as compared with the control and other feed additive groups. The reduced level of cortisol (p 0.05) and elevated levels of progesterone and estradiol (p 0.05) were noted in the Ampbio-fed group as compared to the other feed additive groups. It is therefore concluded that incorporation of Barodon, EM (B. subtilis), and Ampbio in the recommended diet improved the growth and health performance of Korean native heifers.

الوصف: The important platform polysaccharide N-acetylglucosamine (GlcNAc) has great potential to be used in the fields of food, cosmetics, agricultural, pharmaceutical, medicine and biotechnology. This GlcNAc is being produced by traditional methods of environment-unfriendly chemical digestion with strong acids. Therefore, researchers have been paying more attention to enzymatic hydrolysis process for the production of GlcNAc. Hence, in this study, we isolated novel chitinase (Escherichia fergusonii) and chitosanase (Chryseobacterium indologenes, Comamonas koreensis) producing strains from Korean native calves feces, and developed the potential of an eco-friendly microbial progression for GlcNAc production from swollen chitin and chitosan by enzymatic degradation. Maximum chitinase (7.24±0.07U/ml) and chitosanase (8.42±0.09, 8.51±0.25U/ml) enzyme activity were reached in submerged fermentation at an optimal pH of 7.0 and 30°C. In this study, sucrose, yeast extract, (NH4)2SO4, and NaCl were found to be the potential enhancers of exo-chitinase activity and glucose, corn flour, yeast extract, soybean flour, (NH4)2SO4, NH4Cl and K2HPO4 were found to be the potential activator for exo-chitosanase activity. Optimum concentrations of the carbon sources for enhanced chitinase activity were 9.91, 3.21, 9.86, 1.66U/ml and chitosanase activity were 1.63, 1.13, 2.28, 3.71, 9.02, 4.93, and 2.14U/ml. These enzymes efficiently hydrolyzed swollen chitin and chitosan to N-acetylglucosamine were characterized by thin layer chromatography and were further confirmed by high-pressure liquid chromatography. From a commercial perspective, we isolated, optimized and characterized exochitinase from Escherichia fergusonii (HANDI 110) and chitosanase from Chryseobacterium indologenes (HANYOO), and Comamonas koreensis (HANWOO) for the large-scale production of GlcNAc facilitating its potential use in industrial applications.

الوصف: This study hypothesized that dietary feed additive containing probiotics alter either immune-related serum substances or serum metabolites in Hanwoo heifers. A probiotic treatment was given at 0.5% top-dressing of concentrate diet for 6 months. The change of immunological indicators in the blood was analyzed under LPS (Lipopolysaccharide) challenge. One day before administration of LPS, all heifers were fitted with an indwelling jugular vein catheter for serial blood collections. Both a serum tube and an EDTA-coated tube were collected at 30-min intervals from - 2 to 8 hours relative to the LPS challenge at time 0 (1 ㎍/㎏ of BW). Serum was used for analyzing albumin (ALB), glucose (GLU), total protein (TP), triglycerides (TG), phosphorus (IP), and non-esterified fatty acids (NEFA). Plasma was used for analyzing white blood cell (WBC), red blood cell (RBC), platelet (PLT) and inflammation-related factors (NE, LY, MO, EO, BA, Hb, HCT, MCV, MCH, MCHC, RDW, MPV). There were significant differences in ALB, GLU, TG, IP, and NEFA concentration with the passage of hours post challenge (p 0.05). The level of WBC, EO, LY, and MO were reduced after LPS challenge (p > 0.05). In conclusion, LPS challenge after dietary supplementation of probiotics changed the levels of both serum metabolites and inflammation-related factors. The increase of GLU and TG indicated a probiotics-positive response under LPS challenge (p < 0.05).
