Chez la levure S. cerevisiae, le complexe ERMES (ER Mitochondria Encounter Complex) est impliqué dans l'établissement de contacts entre le RE et la mitochondrie. Il est composé de plusieurs protéines membranaires dont Mmm1 dans le RE, Mdm34 et Mdm10 à la membrane externe mitochondriale et d'une protéine soluble Mdm12, qui permet de relier les deux structures. Notre laboratoire a montré que deux composants du complexe ERMES, Mdm34 et Mdm12, étaient ubiquitinés par la ligase d'ubiquitine Rsp5, en particulier, après induction de la mitophagie. Il a également montré que cette ubiquitination était nécessaire pour une mitophagie efficace et qu'un défaut d'ubiquitination du complexe ERMES entraînait une diminution de cette mitophagie. Au cours de ma thèse, j'ai essayé de comprendre comment l'ubiquitination du complexe ERMES pouvait affecter/réguler la mitophagie. De manière générale, nos résultats apportent de nouvelles données concernant les mécanismes moléculaires impliquant ERMES au cours de la mitophagie. ; N the yeast S. cerevisiae, the ERMES complex (ER Mitochondria Encounter Complex) is involved in the establishment of contacts between the ER and the mitochondria. It is composed of several membrane proteins including Mmm1 in the ER, Mdm34 and Mdm10 on the outer mitochondrial membrane and a soluble protein Mdm12, which connects the two structures. Our laboratory has shown that two components of the ERMES complex, Mdm34 and Mdm12, are ubiquitinated by the ubiquitin ligase Rsp5, in particular, after mitophagy induction. They also showed that this ubiquitination is necessary for efficient mitophagy and that a defect of ubiquitination of the ERMES complex reduces mitophagy. During my thesis, I tried to understand how the ubiquitination of the ERMES complex could affect/regulate mitophagy. Overall, our results provide new insights into the molecular mechanism of ERMES action during mitophagy. |
