الوصف: To improve upon current pre-clinical models for cervical cancer, which lack important clinical and pathological features, we aimed to develop a novel, spontaneous, HPV16-expressing carcinoma model that mimics key aspects of HPV-associated carcinogenesis in the female genital tract. Our novel murine model features 1) expression of HPV oncogenes E6 and E7 in the female reproductive tract of mice, 2) spontaneous progression through high-grade squamous intraepithelial lesion (HSIL) to carcinoma, and 3) flexibility to model cancers from different high-risk HPV genotypes. This was accomplished this by injecting plasmids expressing HPV16 E6/E7-Luciferase, AKT, c-myc flanked by inverted terminal repeats, and Sleeping Beauty transposase, into the cervicovaginal tract of C57BL/6 mice followed by electroporation. Cell lines derived from the resultant tumors express HPV16 E6/E7 oncogenes, form tumors in immunocompetent mice and display carcinoma morphology. In sum, we have developed a novel HPV-associated cervicogenital carcinoma model and a novel HPV16 E6/E7 expressing tumor cell line that complement existing murine models of cervical carcinogenesis. Because of its flexibility for modification, our tumor models will be particularly valuable to study oncogene biology and to test candidate therapeutic HPV vaccines or other novel interventions targeting HPV E6/E7-driven tumors.
