    الوصف: Los nucleopoliedrovirus (Baculoviridae) son patógenos letales que infectan de manera natural a las larvas de lepidópteros. El inhibidor de la síntesis de quitina flufenoxuron, ha sido reportado como un potenciador de las propiedades insecticidas de los cuerpos de oclusión (OBs) de nucleopoliedrovirus mediante la degradación de integridad de la matriz peritrófica de las larvas. En el presente estudio se ha examinado el efecto de un tratamiento de flufenoxuron sobre la susceptibilidad de larvas de quinto estadio de Spodoptera exigua (Hübner) y Spodoptera frugiperda (JE Smith) a sus nucleopoliedrovirus homólogos, SeMNPV y SfMNPV, respectivamente. Ensayos preliminares indicaron que una concentración de 1 mg/l de flufenoxuron resultó en la mortalidad de un 6.6 - 13.3% de larvas, el cual podría ser utilizado en combinación con un inóculo de OBs a concentraciones de 1.5 × 104 OBs/ml para SeMNPV o 1.3 × 10⁶ OBs/ml para SfMNPV. El tratamiento de larvas con flufenoxuron no incrementó significativamente la mortalidad por virus en S. exigua (16.7 - 17.2%) o S. frugiperda (28.9 - 34.4%), y no tuvo efecto sobre el promedio de producción de OBs en larvas muertas por virus en ninguna de las especies. La identidad de SeMNPV y SfMNPV fue confirmada para cada virus mediante los perfiles realizados con la digestión con endonucleasas de restricción. Estos hallazgos contrastan con los resultados de estudios previos en los que una concentración más alta de flufenoxuron (100 mg/l) potenciaba la mortalidad por virus en el gusano de seda Bombyx mori L., aunque por razones no muy claras no se reportaba una mortalidad del insecto debida sólo al tratamiento de flufenoxuron.     الوصف: Polyhedrins are viral proteins present in a large family of baculoviruses that form occlusion bodies (polyhedra). These structures protect the virus particles from the outside environment until they are ingested by susceptible insects. Occluded viruses can sustain inclement weather for long periods of time. Therefore, the polyhedra is a natural preservative that keeps the viral structure intact at ambient temperature for years. In a previous study we identified the first 110 amino acids from polyhedrin (PH(1-110)) as a good candidate to carry antigens of interest. As a proof of concept, we produced a fusion protein with PH(1-110) and the green fluorescent protein (PH(1-110)GFP). The fusion protein associates spontaneously during its synthesis resulting in the formation of nanoparticles. Nasal immunization with these nanoparticles and in the absence of any adjuvant, results in a robust immune response with the production of IgG immunoglobulins that remained elevated for months and that selectively recognize the GFP but not PH(1-110). These results indicate that PH(1-110) is poorly immunogenic but capable of enhancing the immune response to GFP.

    الوصف: The occlusion bodies (OBs) of certain alphabaculoviruses are polyhedrin-rich structures that mediate the collective transmission of tens of viral particles to the same insect host. In addition, in multiple nucleopolyhedroviruses, occlusion-derived virions (ODVs) form nucleocapsid aggregates that are delivered to the same host cell. It has been suggested that, by favoring coinfection, this transmission mode promotes evolutionarily stable interactions between different baculovirus variants. To quantify the joint transmission of different variants, we obtained OBs from cells coinfected with two viral constructs, each encoding a different fluorescent reporter, and used them for inoculating Spodoptera exigua larvae. The microscopy analysis of midguts revealed that the two reporter genes were typically segregated into different infection foci, suggesting that ODVs show limited ability to promote the co-transmission of different virus variants to the same host cell. However, a polyhedrin-deficient mutant underwent inter-host transmission by exploiting the OBs of a fully functional virus and re-acquired the lost gene through recombination, demonstrating cellular coinfection. Our results suggest that viral spatial segregation during transmission and primary infection limits interactions between different baculovirus variants, but that these interactions still occur within the cells of infected insects later in infection.

By that, it is considered to be necessaryforOBs purity and qualification when making using formulation.The result of a study undertaken in 2017 showed that theOBs of Spodoptera lituraNPV could be purified by rotational use of two times of SDS and three times of pure water. By the way, the quantity of NPV’s OB could be 2,35 x 1010 OB/ml and their harvested percentage reached 83.93%. The result of study was alsoshowed that it was possible to formulate bioproduct under the wettable powder for a hectare with adosage of 500 gram, in which the concentration of 2,0 x 109 OBs per gram equivalence an using dosage of 1 x 1012 Obs per ha of thecrop. It was also indicated that the efficacy in controllingthe insect pest reached to 81.23% with aproduct without adding 5% Boric axit and 87,14% for the product mixtured with 5% of Boric axit in alaboratory experiment, to 81,13% and 82.92% (equivalent) in greenhouse condition at 7 days after spraying.

    الوصف: Baculoviruses (BV) are highly effective against lepidopteran pests of economic importance such as Spodoptera exigua. The combined use of entomopathogens and macrobiological control agents requires the study of their relationships. Laboratory bioassays were developed to evaluate the interactions between the multiple nucleopolyhedroviruses of S. exigua (SeMNPV) and Autographa californica (AcMNPV), and the predator Chrysoperla carnea. The microscopic examination of predator’s excreta (larval drops and meconia) after the ingestion of BV-infected S. exigua revealed the presence of viral occlusion bodies (OBs). The reinfection of S. exigua larvae with BVs-contaminated excreta by using OBs water suspensions or by direct application both yielded high mortality values but different speed-of-kill results. Meconia killed before in suspensions due to their higher viral load and larval excretion drops did so in direct application due to their liquid nature and their easiness of consumption. The prey-mediated ingestion of SeMNPV and AcMNPV triggered slight effects in C. carnea, which were probably derived from the food nutritional quality. Chrysoperla carnea larvae did not discriminate between healthy and BV-infected S. exigua, while a preference was shown for S. exigua (healthy or infected) vs. Macrosiphum euphorbiae. Our findings present C. carnea, and particularly its larvae, as a promissory candidate for BV dispersion in the field.

    الوصف: The fall armyworm (FAW), Spodoptera frugiperda is the main corn pest in Latin America. As an alternative to chemical insecticide applications, one isolate of Spodoptera frugiperda nucleopolyhedrovirus from soil of an experimental field in Saltillo, Coahuila, Mexico was evaluated. Egg masses of the pest were superficially inoculated by the immersion method, with different nucleopolyhedrovirus concentrations expressed as occlusion bodies/mL (OBs/mL). Artificial diet was also inoculated with different viral concentrations and used to conduct bioassays on first to sixth instar larvae to determine the number of OBs produced per larva. Another assay was performed under greenhouse conditions to evaluate four baculovirus formulations. The use of the immersion method on egg masses caused the highest mortality rates recorded in the larval stage (74.37%), mainly in the first instar, with 64.05% mortality. The number of OBs produced per larva ranged from 5.15 × 106 to 2.3 × 109 , where fifth and sixth instar larvae produced the highest amount. OBs produced by weight unit of larva were registered and higher values were found in the last larval instars. In the baculovirus formulations tested under greenhouse conditions, the mixture with maize flour and starch resulted in significantly increased mortality rates. ; El gusano cogollero (Spodoptera frugiperda) es la principal plaga del maíz en Latinoamérica.     الوصف: Here, we developed a diagnostic ELISA for foot-and-mouth disease using recombinant occlusion bodies (rOBs) of baculovirus. We fused Δ3AB1–3, a polypeptide derived from non-structural proteins of foot-and-mouth disease virus, to polyhedrin (POLH), the major constituent of OBs, under polh promoter. To further assess the most convenient strategy to improve yields, we designed two recombinant baculoviruses, vPOLH and vPOLHE44G. These carried the sequence of the fusion protein POLH-Δ3AB1–3 with an additional copy in cis of polh or polhE44G, respectively, under p10 promoter. Our results show that both viruses expressed POLH-Δ3AB1–3, which was detected by western blot in purified rOBs with anti-POLH and anti-3AB1 antibodies. We also found that vPOLHE44G produced larger polyhedra and a significant increase of antigen yield (p < 0.01). Furthermore, the chimeric protein POLH-Δ3AB1–3 was recognized by sera from experimentally infected animals, showing that translational fusion to POLH does not alter the antigenicity of Δ3AB1–3. Finally, the rOBs were successfully used in an ELISA test to differentiate infected from vaccinated animals.     الوصف: The ability of Baculoviruses to hyper-express very late genes as polyhedrin, the major component of occlusion bodies (OBs) or polyhedra, has allowed the evolution of a system of great utility for biotechnology. The main function of polyhedra in nature is to protect Baculovirus in the environment. The possibility of incorporating foreign proteins into the crystal by fusing them to polyhedrin (POLH) opened novel potential biotechnological uses. In this review, we summarize different applications of Baculovirus chimeric OBs. Basically, the improvement of protein expression and purification with POLH as a fusion partner; the use of recombinant polyhedra as immunogens and antigens, and the incorporation of proteins into polyhedra to improve Baculoviruses as bioinsecticides.     الوصف: Oral infection of insect larvae with baculovirus is an advantageous methodology for producing high levels of recombinant proteins and for achieving plague control. However, many recombinant baculoviruses express a foreign protein in lieu of the polyhedrin and hence do not form occlusion bodies (occ−), resulting in extremely reduced per os infectivity in larvae. To overcome this limitation, stably transformed insect cell lines expressing polyhedrin capable of occluding occ− recombinant baculovirus by trans-complementation were developed to obtain oral inoculum for insect larvae infection. First, the optimum regulatory region of polyhedrin promoter was determined utilizing chloramphenicol acetyl transferase (CAT) as the reporter gene. After infection with occ− baculovirus, the higher expression levels of CAT were achieved when a region of 2735 bp that contained sequences known to have transcriptional enhancer functions were present upstream the polyhedrin coding sequence. This regulatory region was selected to drive polyhedrin expression in insect cell lines. Transfection of Sf9 cells with plasmid carrying polyhedrin gene and stable cell lines established by selection with blasticidin showed polyhedrin expression and, moreover, crystalline polyhedron-like structures were visualized by optic microscopy. Oral infectivity was demonstrated by fluorescence detection in Rachiplusia nu larvae infected with occluded AcGFPpolh− baculovirus obtained using the system presented here. ; Instituto de Biotecnología ; Fil: Lopez, Maria Gabriela.     الوصف: This paper reports studies of thein vitroproduction of a virus fromHelicoverpa armigera(HaSNPV) and its possible use as a specificHelicoverpa/Heliothislarvicide. Growth kinetics ofHelicoverpa zea(H. zea) cells and virus occlusion body yields were compared in three SF900II-based media, namely, SF900II (serum-free), SF900II + 1% serum, or SF900II + 10% serum. Viable cell densities were usually higher in the media supplemented with serum than in the serum-free medium; however, in the serum-free medium, cell diameters were 1.7 times greater (i.e., individual cell volumes were five times larger). Both volumetric production of virus occlusion bodies and production per cell were higher in the serum-free medium than in the media supplemented with serum. However, the infectivity of the occlusion bodies from the serum-free medium was less than that with those from the medium supplemented with 10% serum, when compared in bioassays employing newly hatched larvae. The infectivity of thein vitroproduced occlusion bodies was also less than that ofin vivoproduced occlusion bodies in a commercially available virus product, GemStar. High levels of infection ofH. armigeralarvae obtained in a preliminary field assessment on preflowering tomatoes using thein vitroproduced occlusion bodies indicated the suitability of thein vitroprocess for biopesticide production.
