مستخلص: Halos are circular or ring-shaped cell wall appositions surrounding pathogen entry sites on host leaves. The diameter of halos on infected Blumeria graminis wheat leaves varied between 10 and 100 μm, which reflected specificity of local host plant responses. Using cryoscanning electron microscopy (cryoSEM) without metal sputtering we observed halos as light circles or ring-shaped patterns on the leaves of cereals infected with B. graminis, and on the leaves of several dicotyledonous plants infected with specialized powdery mildews. Hydrogen peroxide treatment of wheat leaves increased the number of cryoSEM detected halos. They were more distinct than in the control without treatment and showed better contrast with surrounding epidermis. The presented cryoSEM method allows simultaneous monitoring of halos and fungal structures and, possibly, visualizes penetration sites with elevated production of reactive oxygen species.
