الوصف: Background: Saliva is a specimen that is easily collected by non-invasive means and does not require well-trained staff; it could be helpful in measuring inflammatory markers to determine COVID-19 severity. The aim of this study was to investigate saliva as an alternative specimen for measuring inflammatory markers IL-10, IL-4, and IL-1β among COVID-19 patients in relation to disease severity. Methods and Results: This cross-sectional study was conducted among COVID-19 patients in a fever clinic, isolated hotels, and hospitals providing care for positive COVID-19 patients and in public health centers for negative control patients in Jeddah, Saudi Arabia. A total of 151 subjects participated in this study, including 101 patients with COVID-19 and 50 healthy controls. Patients with COVID-19 were categorized according to the severity of their symptoms into mild (n=50) and severe cases (n=51). The salivary concentrations of IL-4, IL-10, and IL-1β were measured using sandwich MyBioSource ELISA Test Kits. The age of the study population ranged from 19 to 70 years old, with a mean age of 43.3±13.0 years. The distribution of the study population showed that more of the patients were men (65[64.4%]) than women (36[35.6%]) (P=0.004). The frequency of severe infection in men was higher than in women (35[68.6%] and 16[31.4%], respectively, P=0.008). The group of severe cases was significantly older than the group of mild cases (47.9±11.03 years and 38.64±13.82 years, respectively, P=0.0007). The volume of saliva was the smallest in severe COVID-19, compared to mild cases and controls (P=0.0000 in all cases). The salivary levels of IL-4, IL-10, and IL-1β were greater in the severe cases than in mild cases and controls (46.14±11.61 pg/mL, 12.86±1.99 pg/mL, and 27.45±11.47 pg/mL versus 19.31±5.72 pg/mL, 7.96±2.12 pg/mL, and 6.59±1.90 pg/mL, respectively; P=0.0000 in all cases). The salivary levels of IL-4 and IL-10 in mild cases were greater than in controls (19.31±5.72 pg/mL and 7.96±2.12 pg/mL versus 15.30±4.36 pg/mL and 6.02±0.89 pg/mL, respectively; P=0.0329 and P=0.000, respectively), but salivary IL-1β levels in mild cases did not differ from controls (6.59±1.90 pg/mL vs. 6.03±2.28 pg/mL, P=0.9129). Conclusion: Saliva could be used as an alternative sample in measuring IL-10, IL-4, and IL-1β with the suggestion of using IL-10 and IL-4 as markers for predicting disease severity.

الوصف: We describe a population of the acanthocephalan Corynosoma australe Johnston, 1937 (Polymorphidae) from a California sea lion Zalophus californianus (Lesson, 1828) in California using novel scanning electron microscopy (SEM) images, Energy Dispersive x-ray analysis (EDXA), and molecular analysis for the first time. The taxonomic history of C. australe is replete with accounts using only line drawings some of which proved erroneous. The distribution of ventral spines on the female trunk has been the primary distinction between C. australe and Corynosoma obtuscens Lincicome, 1943, its junior synonym; being continuous in the latter but discontinuous posteriorly in the former species. The distribution of ventral spines is invariably discontinuous in males. Our redescription and SEM images help to resolve this issue further validating the synonymy. Morphological variability has been documented between our California population and others from various host species in California, South Australia, South Shetlands, and the Argentinian coast. Our SEM images document features not previously detectable in line drawings, erroneously reported or missed in previous accounts. The EDXA spectra show high levels of calcium and phosphorous and low levels of sulfur characteristic of C. australe. EDXA for other species of Corynosoma Lühe, 1904 provide support for the diagnostic distinction of C. australe. EDXA spectra were shown to be species specific and have diagnostic value in the taxonomy of the Acanthocephala. Our molecular analysis used amplification of 18S of ribosomal DNA and cytochrome c oxidase 1 (Cox1) gene. Phylogenetic analyses for Cox1 gene revealed a close relationship between Corynosoma hannae Zdzitowiecki, 1984 and C. australe. The phylogenetic trees confirmed that the isolates belonged to C. australe. The haplotype network inferred by Cox1 with C. australe sequences revealed that haplotypes clearly separated from each other and formed clusters related to samples from the Northern Hemisphere (the USA and Mexico), and the second from the Southern Hemisphere (Argentina, Brazil and Peru).

الوصف: Tyrosine-containing pharmaceuticals’ (TPh) potential to inhibit SARS CoV-2 3-chymotrypsin-like proteases (3CLpro) and nonstructural protein 16 (NSP16) has been explored using docking studies, molecular dynamics simulations, and density functional theory. The TPh with FDA approval showed excellent contact with the active site pockets of 3CLpro and NSP16. Their binding affinity scores ranged from −5.8 to −4.9 kcal/mol and −6.3 to −4.8 for 3CLpro and NSP16, respectively. A 100-ns molecular dynamics simulation confirmed the stability of the carbidopa/NSP16 complex and N-acetyl tyrosine with both target enzymes. Further, the HOMO-LUMO transitions, molecular orbitals, and dipole moments of carbidopa, droxidopa, and N-acetyl tyrosine were computed using density functional theory (DFT). Considering N-acetyl tyrosine and carbidopa’s substantial inhibitory activity, it is recommended to investigate them further in order to explore their application for the treatment of COVID-19 or any other coronaviruses in the future.

الوصف: Water contamination is one of humanity's threatening problems. This study synthesized pristine carbon nanotubes (PCNTs) from commercial gasoline (CG) as a relatively low-cost precursor. 200 mL of CG yielded an average CNTs mass of 26.23 ± 0.97 g. The functionalized carbon nanotubes (FCNTs) were produced by acid-treatment of PCNTs, and the yield was 93.3%. The surface area of PCNTs and FCNTs was 29.69 and 66.16 m2 g−1. The FTIR and EDX indicated an introduction of oxygenated groups on the FCNTs. The FCNTs removed ciprofloxacin (CIP) and indigo carmine (IC) from water with an adsorption capacity of 95.5 and 93.0 mg g−1, respectively, while the PCNTs showed 78.9 and 63.7 mg g−1, respectively. The pH 4.0 and 6.0 suited the adsorption of CIP and IC on both sorbents. The adsorption processes for all sorbent-sorbate systems were spontaneous and exothermic. The FCNTs and PCNTs recyclability showed an average efficiency of 90.0%.

الوصف: This paper explores a dual inhibition of main protease (Mpro) and nonstructural protein 10/nonstructural protein 16 (NSP16) methyltransferase complex as the key targets for COVID-19 therapy. These are based on the new Schiff-base ligand that was obtained from the condensation of (4-chloro-3-methyl phenyl) hydrazine with 2-pyridine-carboxaldehyde and its novel Schiff-base metal complexes. These include Ni(II), Pd(II), Pt(II), Zn(II), and Hg(II). The newly synthesized compounds have been characterized using FT-IR, 1H NMR, 13C NMR, and elemental analysis. The results suggested that the Schiff-base ligand is coordinated as a bidentate ligand through the nitrogen atoms of the azomethine group and pyridyl ring. In addition, the biological activity of the prepared complexes was examined against Pseudomonas aeruginosa and Staphylococcus aureus, and the results showed that the Zn(II) complex has the highest activity compared with other compounds. The active sites were found by looking at the molecular electrostatic potential (MEP) maps of the above ligands and complexes. The activity of the compound and its Ni(II) and Zn(II) complexes against Mpro and NSP10/ NSP16 was investigated using a molecular docking approach. They showed excellent binding energies ranging from −5.9 to −7.2 kcal/mol and −5.8 to −7.2 for Mpro and NSP16, respectively. All conformers of the metal complexes were docked with the active site of the NSP16 receptor, showing a binding affinity of 100%. According to our knowledge, this was the first report of these metal complexes as dual inhibitors for Mpro and NSP16 of SARS-CoV-2.

الوصف: Medicinal herbs have long been utilized to treat various diseases or to relieve the symptoms of some ailments for extended periods. The present investigation demonstrates the phytochemical profile, molecular docking, anti-Candida activity, and anti-viral activity of the Saussurea costus acetic acid extract. GC-MS analysis of the extract revealed the presence of 69 chemical compounds. The chemical compounds were alkaloids (4%), terpenoids (79%), phenolic compounds (4%), hydrocarbons (7%), and sterols (6%). Molecular docking was used to study the inhibitory activity of 69 identified compounds against SARS-CoV-2. In total, 12 out of 69 compounds were found to have active properties exhibiting SARS-CoV-2 inhibition. The binding scores of these molecules were significantly low, ranging from −7.8 to −5.6 kcal/mol. The interaction of oxatricyclo [20.8.0.0(7,16)] triaconta-1(22),7(16),9,13,23,29-hexaene with the active site is more efficient. Furthermore, the extract exhibited significant antimicrobial activity (in vitro) against Candida albicans, which was the most susceptible microorganism, followed by Bacillus cereus, Salmonella enterica, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa, respectively. On the other hand, its antiviral activity was evaluated against HSV-1 and SARS-CoV-2, and the results showed a significant positive influence against HSV-1 (EC50 = 82.6 g/mL; CC50 = 162.9 g/mL; selectivity index = 1.9). In spite of this, no impact could be observed in terms of inhibiting the entry of SARS-CoV-2 in vitro.

الوصف: Topologically protected magnetic textures are promising candidates for information carriers in future memory devices, as they can be efficiently propelled at very high velocities using current-induced spin torques. These textures—nanoscale whirls in the magnetic order—include skyrmions, half-skyrmions (merons) and their antiparticles. Antiferromagnets have been shown to host versions of these textures that have high potential for terahertz dynamics, deflection-free motion and improved size scaling due to the absence of stray field. Here we show that topological spin textures, merons and antimerons, can be generated at room temperature and reversibly moved using electrical pulses in thin-film CuMnAs, a semimetallic antiferromagnet that is a testbed system for spintronic applications. The merons and antimerons are localized on 180° domain walls, and move in the direction of the current pulses. The electrical generation and manipulation of antiferromagnetic merons is a crucial step towards realizing the full potential of antiferromagnetic thin films as active components in high-density, high-speed magnetic memory devices.
