الوصف: 本研究之目的乃在分離純化枯草菌及木瓜蛋白酶粗酵素製品所含 幾丁聚醣酶(chitosanase, EC 3.2.1.132),並探討其生化學性質與應用。 枯草菌粗酵素製品分離之幾丁聚醣酶其水解幾丁聚醣之最適pH 為 4.0,最適溫度為50℃,Km 值為1.3 mg/mL,以等電焦集電泳測得等 電點為3.8,膠體過濾法測得分子量為27.4 kDa,重金屬離子 Hg2+(2.5mM)及Ag+(2.5mM)顯著抑制酵素活性。木瓜酶粗酵素製品純 化之幾丁聚醣酶其水解幾丁聚醣之最適pH 為4.6,最適溫度為70℃, Km 值為1.58 mg/mL,對熱頗穩定(70℃保溫60 分鐘尚有80%活性), 以膠體過濾法測得分子量為12.4 kDa,水解幾丁聚醣之主要產物為低 分子量幾丁聚醣(∼10kDa)及若干幾丁寡醣。 ; The purpose of this study is to purify and characterize chitosanases from crude enzyme preparations of Bacillus subtilis and papaya latex. Chitosanase isolated from a crude enzyme preparation of Bacillus subtilis had an optimal pH of 4.0, an optimal temperature of 50℃ and a Km of 1.3 mg/mL for chitosan hydrolysis. The isoelectric point was 3.8, as estimated by isoelectric focusing electrophoresis. The molecular mass was 27.4 kDa, as estimated by gel filtration. Heavy metal ions of Hg2+(2.5 mM) and Ag+(2.5 mM) significantly inhibited the activity of the enzyme. Chitosanase isolated from a crude enzyme preparation of papaya latex had an optimal pH of 4.6, an optimal temperature of 70℃ and a Km of 1.58 mg/mL for chitosan hydrolysis. The enzyme was thermostable (80% of the activity remained after incubation at 70℃ for 60 min). The molecular mass was 12.4 kDa, as estimated by gel filtration. The main end product of the chitosan hydrolysis catalyzed by the enzyme was low molecular weight chitosan (∼10 kDa) and some chitooligosaccharides.
