مستخلص: To investigate the effect of transforming growth factor - ß 1 siRNA neural stem cells transplantation on the recovery of spinal cord injury in rats, and analyze its mechanism. The expression of TGF - ß 1 protein was detected by Western blot. 60 adult female SD rats were selected to establish the acute spinal cord injury model. The rats were randomly divided into two groups, 30 rats in each group. NSCs were transplanted in the control group and TGF - ß 1-siRNA in the observation group. The expression of TGF - ß 1 mRNA, protein, synaptophysin and HRP retrograde tracing were compared between the two groups. Compared with the control group, the lower limb function score of the observation group increased significantly, the difference was statistically significant (P < 0.05). Compared with the control group, TGF - ß 1 mRNA transcription and protein expression in the observation group increased significantly (P < 0.05). Compared with the control group, the expression level of synaptophysin in the observation group increased significantly (P < 0.05). The number of HRP positive nerve fibers and cm DIL positive cells were significantly increased in the observation group compared with the control group (P < 0.05). TGF - ß 1-siRNA neural stem cells can promote the recovery of spinal cord injury in rats, participate in the recovery of lower limb motor function, and TGF - ß has a negative correlation with the recovery of nerve function, providing new experimental data and evidence for the treatment of spinal cord injury.
