الوصف: In the research, SACCHACHITIN (SC) is component containing polysaccharide and chitin derived from fruiting body of Ganoderma tsugae was proved to be a bio-compatible and biodegradable material. Through extracted by hot water and alcohol, the residue was reach up to 90% and soluble in water was poor. In order to solve this problem, we developed the chemical treatment by SiliaCat-TEMPO mediated oxidation of SC with Sodium bromide and Sodium hypochlorite system. These nanoscale SC materials were characterized by FT-IR, solid state NMR, X-ray diffraction, carboxylate content, size, and zeta potential for the chemical properties. The physical properties were also evaluated by gel forming properties and water retention properties. The study showed that the nanoscale SiliaCat-TEMPO-oxidation of SC oxidized by1.6, 5.0, and 10.0 mmol-NaClO/g-SC, were named as ST016SC, ST050SC, and ST100SC. With the increasing of the oxidation, the SiliaCat-TEMPO oxidized SC by FT-IR showed a sharp peak at 1620 cm-1 increased respectively which was the C=O stretching. The result in chemical properties with carboxylate content showed increasing of the oxidation, and could be more soluble to become greatest biomaterials After lyophilizing, all samples at 2% (w/v) concentration, they presented high porosity structure. The results of MTT assay of WS-1 human fibroblast and MC3T3E1 cell viability showed no cytotoxicity and high biocompatibility. All the results indicated that these biomaterials have great potential to be conducted to animal study more safely.
