An in vitro, working model of the cholera - gut system was developed. The model was patterned after a TSH - thyroid system that this author had developed in past years. A duodenal homogenate was challenged with cholera toxin in the presence of [14C] labeled adenosine-5'-triphosphate (ATP) and/or [14C] labeled NAD with the appropriate controls. The system was then incubated for eight hours. Cyclic AMP, ATP and NAD were isolated from the homogenate via High Performance Liquid Chromatography (HPLC) methods and radioactivity evaluated by scintillation methods. |
