الوصف: Spontaneous regeneration of osteochondral defects (OD) with hyaline cartilage is impossible due to tissue avascularity and small cellular number. Therefore, cartilage trauma often results in degenerative processes of osteoarthritis, affecting joint mobility and the patient’s quality of life. Many therapeutic techniques, like microfracture and autologous chondrocyte implantation, have been applied but with many disadvantages, such as increased morbidity. Regenerative medicine has proposed the application of stem cells (SCs) for cartilage healing. Veterinary literature includes several studies about SCs usage, but most of them lack control groups, population homogeneity and a randomized, double-blinded design. Also, because of different protocols, duration and postoperative therapy, the comparison between the therapeutic properties of SCs is impossible. This randomized, controlled, double-blind study aimed to evaluate the osteochondrogenic potentials of rabbit allogeneic stem cells derived from adipose tissue (ASCs), bone marrow (BMSCs) and synovial membrane (SMSCs) encapsulated in fibrin glue in vivo. In addition, this study investigated the chondrogenic properties of fibrin glue in critical-sized osteochondral defects. A 3x3 mm-sized osteochondral defect was created in the femoral patellar on both knees of 48 healthy New Zealand rabbits, except from the left knees of 8 rabbits (control group) in which the osteochondral defects were 2x3mm. The rabbits were randomly divided into four groups: control (untreated) group (n=8), fibrin glue/fibrin glue with ACSs group (n=16), fibrin glue/fibrin glue with BMSCSs group (n=16), fibrin glue/fibrin glue with SDSCs group (n=16). After 12 weeks a macroscopic evaluation and a histology assessment were performed according to the International Cartilage Repair Society and O’Driscoll scale, accordingly. Also, immunohistochemical analysis of collagen II and aggrecan was completed. Finally, lactate dehydrogenase (LDH) was measured in the rabbits’ blood and synovial fluid. ...
