مصطلحات موضوعية: Bile diversion surgery, used for treating obesity, increases the circulating bile acids (BAs) and reduces food and cocaine reward-related behavior in mice. Feeding mice addicted to cocaine with obeticholic acid (OCA), a BA analog, induced the same effect of the surgery. The interactions of OCA with DAT heterologously expressed in X. laevis oocytes showed that OCA promotes a transient Na+-inward current, without affecting dopamine (DA) affinity and kinetic parameters. Docking simulations suggest that OCA could stabilize DAT in an occluded conformation (Romanazzi et al., 2021). The second treatment with OCA does not elicit any current. To fully recover the current elicited by OCA, it is necessary to activate the maximal transport current with substrate at saturating concentration. Reducing DA concentration to a value proximal to the K0.5, induces a smaller OCA current. The same happens with the transport of norepinephrine or serotonin, which are also DAT substrates with a lower affinity than DA. These experiments confirm the model obtained by molecular docking, suggesting that OCA binds, opens a transient conductance, and then freezes DAT in an occluded conformation that makes it resistant to other BA-mediated alterations of the membrane potential. DAT occluded state can be recovered only in the presence of the physiological substrates. Testing the BAs on other SLC6 will shed a light on the specificity of the interaction.
