    الوصف: Introduction: Elevated LDH levels have been extensively reported as a biomarker of poorer outcome in patients with melanoma during the chemotherapeutic era. The role of LDH level as a prognostic factor for treatment outcomes in patients with metastatic melanoma treated with immunooncological therapy has also been reported but requires further analysis. Objectives: In this study, we aimed to evaluate the prognostic value of lactate dehydrogenase (LDH) among patients with metastatic and unresectable melanoma treated with pembrolizumab in terms of pro- gression-free survival (PFS). Methods: The study included 59 patients with unresectable and met- astatic melanoma treated with pembrolizumab. A comparison was performed between patients with normal and elevated levels of LDH in terms of PFS, with subgroup analysis. Results: There was a significant reduction in PFS among patients with elevated levels of LDH compared with patients with normal levels of LDH (NR vs. 5 months; P=0.02). Patients with elevated LDH levels were older (P=0.01), with liver metastasis (P=0.004), and with less frequent CNS deposits (P=0.028). Conclusion: Although novel agents improved outcomes in patients with melanoma, high levels of LDH persist as an independent prog- nostic biomarker of poor prognosis.

    الوصف: Istraživanje je provedeno radi utvrđivanja učinaka suplementacije omega 3 masnim kiselinama na razinu oksidativnog stresa, mišićnog oštećenja i upalnih biljega kao akutnog odgovora na trening s opterećenjem u mladih sportaša. U okviru dvostruko slijepog dizajna eksperimenta sa slučajnim odabirom dvadeset je ispitanika bilo podijeljeno u dvije jednake grupe u kojima je svaki ispitanik uzimao dnevno tri kapsule (3000 mg) omega 3 masnih kiselina ili, u kontrolnoj grupi, placebo kapsule tijekom sedam dana. Svi ispitanici su podvrgnuti visokointenzivnom treningu s opterećenjem. Venski krvni uzorci bili su prikupljeni tjedan dana prije treninga, neposredno prije treninga te 24 sata nakon vježbanja. Mjerene su serumske vrijednosti malondialdehida (MDA), antioksidacijskog kapaciteta u plazmi (FRAP), C reaktivnog proteina (CRP), kreatin kinaze (CK) i laktat dehidrogenaze (LDH). Koncentracije MDA, CRP i CK bile su statistički značajno veće 24 sata nakon vježbanja u placebo grupi nego li u grupi koja je uzimala omega 3 masne kiseline (p=0,005). Prosječne vrijednosti ukupnog antioksidacijskog kapaciteta u obje grupe nisu pokazale značajne razlike između mjerenja neposredno prije vježbanja i 24 sata nakon vježbanja (p>0,005). Aktivnost LDH bila je statistički značajno viša 24 sata nakon vježbanja u obje grupe (p<0,05). Rezultati ovog istraživanja pokazuju da visokointenzivne vježbe s opterećenjem izazivaju oksidativni stres, sistemske upalne procese i povećavaju razinu pokazatelja staničnih oštećenja u sportaša.     الوصف: U radu je ispitan utjecaj osmoze na ekspresiju gena i aktivnost glavnih enzima koji sudjeluju u fermentaciji zrelih zelenih rajčica (Solanum lycopersicum cv. Ailsa Craig), i to uranjanjem plodova u vodu i rasol što sadržava 5 ili 10 % NaCl. Površina je plodova prije obrade sterilizirana da bi se spriječio rast mikroorganizama na pokožici ploda. Analizirani su plodovi rajčice i rasol tijekom 0,5; 1; 1,5; 2; 3; 7 i 14 dana skladištenja, te ispitani ovi parametri: promjena ekspresije gena i aktivnost enzima koji sudjeluju u fermentaciji, te fizikalno-kemijska svojstva plodova (udio topljivih tvari, titracijska kiselost, pH-vrijednost i čvrstoća). Utvrđene su razlike u dobivenim rezultatima, i to na molekularnoj razini te u kakvoći plodova. Zaključeno je da utjecaj fermentacije na tkivo ploda rajčice ovisi o složenoj kinetici prelaska otopljenih tvari iz plodova u otopinu zbog razlike osmotskih tlakova. Skladištenje u vodi uzrokovalo je pucanje plodova koji su imali veću titracijsku kiselost i manji udio topljivih tvari. Ekspresija gena i aktivnost enzima pokazali su da je došlo do povećanja anaerobnog metabolizma u tim plodovima, u usporedbi s onima skladištenim u rasolu. Skladištenje plodova u izotoničnoj otopini (5 % soli) nije uzrokovalo njihovo pucanje, a u hipertoničnoj (10 % soli) otopini nije došlo do pucanja već do dehidracije plodova. Dodatkom soli smanjen je intenzitet fermentacije, što je dovelo do manje ekspresije gena i aktivnosti enzima. Trajnost plodova skladištenih u izotoničnoj otopini bila je veća od onih skladištenih u vodi ili hipertoničnoj otopini. Manja koncentracija soli u otopini nije bitno utjecala na metabolizam fermentacije te kakvoću plodova, a spriječila je njihovo propadanje tijekom skladištenja. ; The effect of osmotic strength on gene expression and activity of the major enzymes of fermentative metabolism of mature green tomato fruit (Solanum lycopersicum cv.     الوصف: Although there is no routine procedure for determination of serum markers in patients with malignant melanoma (MM), some markers are being studied as potentially useful prognostic tools. Serum lactate dehydrogenase (LDH), protein S-100B, melanoma-inhibiting activity (MIA) and tyrosinase may correlate with melanoma progression. In this study, the results of determination of S100 protein, LDH, MIA and tyrosinase in the serum of 50 patients with MM (stages I-IV) were determined. The increased values of MIA were found in 26% patients in stage I, while in 50% patients in stage IV. Increased S-100 protein was found in 13% patients in stage I while in 50% patients in stage IV. The increased values of LDH were found in 26% patients in stage I, while in 25% patients in stage IV. The positive serum tyrosinase was noticed in 17.3% patients in stage II, while in 25% patients in stage IV. The obtained results have revealed no significant differences between the groups in higher and lower stages of the disease, indicating that blood markers are not reliable prognostic factors for MM progression.

    الوصف: Istraživani su učinci karbamatnog insekticida metomila u štakora. Nakon jednokratne peroralne doze od 3, 5 ili 7 mg/kg nađeno je značajno povećanje sveukupne aktivnosti laktat dehidrogenaze (LDH) nakon prvog dana tretmana. Najveća je aktivnost utvrđena trećeg dana nakon tretmana metomilom u dozi od 7 mg/kg. Enzimska aktivnost postepeno se smanjivala i sedmoga se dana normalizirala. Posebno je zapaženo povećanje aktivnosti izoenzima LDH-3 i LDH-4. U posebnim je pokusima utvrđeno značajno smanjenje težine slezene i vijabilnosti splenocita u štakora tretiranih sa 6 mg/kg i 8 mg/kg metomila i to nakon prvog i nakon trećeg dana tretmana. Toksični učinci na slezenu mogli su se spriječiti pretretiranjem sa 60 mg/kg N-acetilcisteina. Prema rezultatima ovih pokusa čini se da bi toksični učinak metomila na slezenu mogao biti izravan učinak metomila na stanice slezene a ne putem kolinergičnog mehanizma. Važnost ovakvog citotoksičnog učinka i mehanizme citotoksičnosti i njihove povezanosti s reaktivnim oksidativnim procesima pri staničnom oštećenju valja još istraživati. ; The toxic effect of methomyl was studied in rats after a single or repeated oral administration. Rats treated with a single dose of methomyl (3, 5, or 7 mg/kg) showed significant increase (P<0.05) in total lactate dehydrogenase activity on day 1. The highest level of LDH activity was observed on day 3 in rats receiving 7 mg/kg of methomyl. The total LDH activity returned to normal on day 7 after dosing. Specific increases in LDH-3 and LDH-4 isoenzyme activities were observed. In rats treated with a single dose of 6 and 8 mg/kg of methomyl, spleen weight and splenocyte viability significantly dropped (P<0.05) on days 1 and 3, respectively. Splenotoxicity was prevented by pretreatment with 60 mg/kg of N-acetylcysteine. The results suggest that the splenotoxic effect of methomyl is more likely directly related to oxidative cell injury than to cholinesterase inhibition.
